冠心平对急性缺血犬心肌细胞凋亡及Bcl-2、Bax蛋白表达的影响(1)

     摘 要:目的:研究冠心平对冠脉结扎导致心肌缺血犬心肌细胞凋亡及Bcl-2、Bax蛋白表达的影响。方法:36头Beagle犬,分成假手术组,心肌缺血模型组,心可舒组,小、中及大剂量冠心平组。前两组分别给予空白胶囊,其他各组每日两次口服相应剂量的药物,连续给药10天,10天后进行冠状动脉结扎实验。结扎180min后取心肌切片保存,检测心肌细胞凋亡指数(AI),并用免疫组化法检测心肌组织Bcl-2、Bax蛋白的表达。结果:与模型组相比,心可舒组及冠心平小、中、大剂量组的心肌凋亡指数水平均有所下降,Bcl-2蛋白表达增强,Bax蛋白表达减少,与模型组比较均具有统计学意义(P<0.01)。结论:冠心平具有减轻缺血心肌细胞凋亡,提高Bcl-2蛋白表达,降低Bax蛋白表达,增加Bcl-2/Bax比值的作用。提示冠心平对缺血犬具有抑制心肌细胞凋亡,减轻细胞凋亡对缺血心肌的损伤的作用。

    The Impact of Guanxinping on the Cardiocytes Apoptosis and the Expression of

    Bcl-2 and Bax in the Acute Cardiac Ischemic Canine Model

    Abstract:Obsjective:To investigate the impact of Guanxinping on the cardiocytes apoptosis and the expression of Bcl-2 and Bax in the cardiac ischemic canine model.Methods:36 canine dogs were assigned into sham procedure group, cardiac ischemic group, xinkeshu group, and small dose of Guanxinping,median dose of Guanxinping and large dose of Guanxinping group. Before the procedure, the groups were pretreated with control blank capsules, blank capsules, xinkeshu, small dose of Guanxinping, median dose of Guanxinping and large dose of Guanxinping, respectively, for ten days. The cardiac ischemic model was created by ligating a coronary artery, while the sham dogs with the same artery isolated but not ligated. After 180 minutes of ligating, the cardiac tissues were collected from the ligated area. The apoptosis index(AI) of the cardiac myocytes was determined and the expression of the Bcl-2 and Bax were measured by immunohistochemistry.Results:Compared with the control group, the AI of the xinkeshu group, small dose Guanxinping group, median dose of Guanxinping group and large dose of Guanxinping group significantly decreased, while the expression of Bcl-2 and Bax increased ......
