5种中药注射剂对多柔比星所致心肌H9c2损伤保护作用(1)

     摘要:目的:考察5种中药注射剂对多柔比星(DOX)作用下H9c2心肌细胞损伤的保护作用。方法:应用H9c2心肌细胞筛查体系,通过检测细胞存活率、过氧化物岐化酶(SOD)活性和氧化代谢产物(MDA)含量,考察参麦注射剂、丹参注射剂、冠心宁注射剂、香丹注射剂和黄芪注射剂等5种中药注射剂对暴露于1.7×10-4至2.2×10-9 M浓度DOX下H9c2心肌细胞的存活率的影响。结果:黄芪注射剂和香丹注射剂能提高DOX作用下心肌细胞的存活率,有效地缓解DOX所引起的心肌细胞MDA浓度增高,提高SOD酶活性且不影响DOX的体外抗肿瘤活性。结论:黄芪注射剂和香丹注射剂能降低DOX对心肌细胞的毒性作用,可能是通过增加抗氧化物酶活力,减少氧化产物生成,从而减轻氧化应激损伤这些途径实现的。

    Protective Effect of Five Injections on Doxorubicin-Induced

    Cytotoxicity in Cardiac Myoblasts H9c2

    Abstract:Objective:To test whether five injections can transfer its antioxidant function into the protection of the doxorubicin (DOX)-induced cardiotoxicity in cardiac myoblasts H9c2. Method:H9c2 cells were exposed to DOX and/or five injection. The peak concentration of five injection of protective effect on DOX-induce was evaluated through examination, level of malondialdehyde (MDA) and superoxide dismutase (SOD) activity.The effects of five injections on the cytotoxicity of DOX to H9c2 cells and A549 lung carcinoma cells were studied. Results:Huangqi injection and Xiangdan injection had protective effect on H9c2 cells against DOX and significantly increased SOD activity and decreased the MDA level in H9c2 experimental system damaged by DOX exposure while not attenuate the antitumor activity of DOX on A549 carcinoma cells. Conclusion:Our evidence indicates that Huangqi injection and Xiangdan injection elicited a typical protective effect on DOX-induced cytotoxicity in cardiac myoblasts H9c2 via suppressing oxidative stress.
