洋金花提取物对肠缺血再灌注大鼠的作用研究(1)

     摘 要:目的:研究洋金花提取物对大鼠缺血再灌注期间肠黏膜的保护作用及机制。方法:采用大鼠肠缺血再灌注模型,大鼠分为假手术组、模型组、阳性对照组、给药组。用Chiu氏评分法观察肠黏膜的损伤程度并检测肠黏膜中肿瘤坏死因子(TNF-α)和白细胞介素(IL-6)。结果:与假手术组比较,模型组可见明显的肠黏膜损伤,TNF-α和IL-6水平明显升高;与模型组比较,给药组肠黏膜病理损伤程度明显减轻,且TNF-α和IL-6水平明显降低。结论:洋金花提取物对大鼠肠缺血再灌注后的肠道具有较好的保护作用,且该作用与TNF-α和IL-6的减少有关。

    Research on Effect of Extraction From Flos Daturae on

    Rats with Intestinal Ischemia-Reperfusion

    Abstract:Objective:To investigate the protective effects and mechanism of extraction from Flos Daturae on rats with ischemia/reperfusion injury. Methods:By using rat model of intesinal I/R injury,Male SD rats were divided into sham-operation group,control group,positive control group and treatment group. Chiu's count was used to assess the changes in intestinal pathological morphplogy,and the contents of tumor necrosis factorα(TNF-α),interleukin-6(IL-6) in intestinal mucosa were measured. Results:The intestinal mucosa was more clearly in control group compared to those in sham-operation group,and TNF-α and IL-6 contents in control group were significantly increased inversely,the intestinal mucosa was reduced significantly in treatment group compared to those in control group,and TNF-α and IL-6 contents in treatment group were significantly decreased. Conclusion:Pre-treatment with extraction from Flos Daturae has protective effect on intestinal ischemia-reperfusion injury in rats,which may be caused by decreased contents of TNF-α and IL-6.

    肠缺血/再灌注损伤( ischemia reperfusion injury, I/R)是常见的组织器官损伤之一,亦是造成远隔重要脏器(肺、心、肾)损伤的“启动器官”,研究表明,肠缺血/再灌注损伤不仅会引起局部组织损害,黏膜屏障功能受损,导致肠内细菌和毒素移位到体循环,引起网状内皮系统发生一系列反应,进而导致大量相关介质和细胞因子的释放,诱发全身炎性反应综合征(SIRS)和多器官功能不全综合征(MODS) ,因而肠缺血再灌注损伤越来越受到重视,其发生发展机制及防治措施的探索也成为近年来研究的重点[1-2]。

    洋金花为茄科植物白曼陀罗Datura metel L.的干燥花,性辛,温;有毒。具有平喘止咳、镇痛、解痉等功效,临床上常用于咳嗽哮喘、脘腹冷痛、风湿麻痹、小儿慢惊、外科麻醉等症状[3]。现有的研究表明洋金花中主要成分是以东莨菪碱和阿托品为主的生物碱类物质,该生物碱类物质作用于中枢系统具有显著的镇静效果,作用于周围神经系统能够解除血管痉挛 ......
