扶正固本中药对急性肺损伤大鼠免疫调节作用研究(1)

     摘 要:目的:探讨扶正固本中药对内毒素所致大鼠急性肺损伤(Acute Lung Injury)的免疫调节作用。 方法:30只SD雄性大鼠随机分成5组:正常对照(NS)组、急性肺损伤(ALI)组、黄芪(HQ)组、参附(SF)组、黄芪+参附(HQ+SF)组。NS组、ALI组予以生理盐水(NS)腹腔注射,HQ组予以黄芪注射液腹腔注射,SF组予以参附注射液腹腔注射,HQ+SF组予以黄芪注射液+参附注射液腹腔注射。给药7天后除正常对照组外均尾静脉注射内毒素。分别测定各组大鼠支气管肺泡灌洗液(BALF)及血液中干扰素-γ(IFN-γ)、白介素-4(IL-4)浓度。同时观察各组大鼠肺组织的病理改变。结果:(1)与正常对照组比较,ALI组大鼠BALF及血液中IFN-γ含量明显减少(P<0.01),IL-4含量明显增加(P<0.01)。与ALI伤组比较,HQ组IFN-γ含量明显增加(P<0.01),SF组IFN-γ含量无明显变化(P>0.05),HQ+SF组IFN-γ含量较ALI组、HQ组、SF组明显增加(P<0.01)。而各组IL-4含量较ALI组无明显变化(P>0.05)。(2)与ALI组相比,HQ组、SF组、HQ+SF组的肺系数、BALF中的蛋白含量均低于ALI组,其中HQ+SF组下降更为明显。结论:黄芪、参附、黄芪加参附中药液对ALI均有防治作用。其中黄芪、黄芪加参附中药液通过调节炎症反应而发挥对ALI的防治作用,对 ALI以促炎症反应为主时可能更为有益;参附对抗炎症介质和促炎症介质均无明显影响,但肺病理结果显示肺损伤程度明显减轻,其对ALI的防治作用可能通过其他途径实现。

    Immunity Control Action to Rats of Acute Lung Injury by Traditional Chinese Medicine

    Abstract:Objective:To study protection mechanisms of traditional Chinese medicine that can tonify vital Qi and consolidate root on acute lung injury rats caused by endotoxin.Methods: Thirty SD rats were divided into five groups: control group,acute lung injury(ALI) group ,Huangqi group,Shenfu group, Huangqi and Shenfu group. Normal saline was injected through abdomen in control group and ALI group, Huangqi injection water was injected through abdomen in Huangqi group, Shenfu injection water was injected through abdomen in Shenfu group, Huangqi and Shenfu injection water was injected through abdomen in Huangqi and Shenfu group. After 7 days, LPS was injected through tail vein in all groups except control group. IFN-γ、IL-4 in the BALF and blood in the five groups of rats were measured. And lung tissue was also used for pathological examination. Results:(1)Compared with the control group, IFN-γ in the BALF and blood was obviously decreased (P<0.01) whereas IL-4 inreased(P<0.01) in ALl group ......
