参麦注射液对心衰犬内源性洋地黄样物质组织浓度的影响(1)

     摘要:目的 考察参麦注射液对心衰犬组织内源性洋地黄样物质(EDLS)浓度的影响。方法 将造模成功4周后的心衰犬6只,随机分为对照组和参麦注射液组,每组3只。对照组予生理盐水20 mL,参麦注射液组予参麦注射液1.551 mL/kg(用生理盐水稀释至20 mL)静脉注射。给药后4 h处死实验犬,分别摘取分离心肌、肾上腺和下丘脑,制成生理盐水匀浆冰冻保存(-80 ℃),用放射免疫分析法检测匀浆中的EDLS浓度。结果 参麦注射液组肾上腺、下丘脑的EDLS浓度高于对照组,心肌EDLS浓度有高于对照组的趋势。结论 参麦注射液可增加心衰犬肾上腺、下丘脑、心肌EDLS浓度。

    The Influence of Shenmai Injection on Tissue Concentration of Endogenous Digitalis-like Substance in Dogs with Heart Failure

    Abstract:Objective To investigate effect of Shenmai injection (SMI) on tissue concentration of endogenous digitalis-like substance (EDLS) in the model dogs with heart failure(HF).Methods Six model dogs with HF were randomly divided into two groups:Control group (n=3) received intravenous injection of normal saline 20 mL and SMI group (n=3) received intravenous injection of SMI(1.551 mL/kg).All dogs were executed at 4 h after administration.The homogenate of samples of heart,adrenal gland and hypothalamus were frozen (at -80 ℃).The EDLS concentrations in tissue samples were determined by radio-immunity assay.Results EDLS concentration of adrenal gland and hypothalamus in SMI group were significantly higher than that in control group.Conclusion SMI could increase the EDLS concentration of adrenal gland,hypothalamus and heart in the model dogs with HF.

    内源性洋地黄样物质(endogenous digitalis-like substance,EDLS)是近年来在人和哺乳动物体内发现的一种具有Na+-K+-ATP酶活性的内源性物质,有强大的利尿、强心作用[1],与心衰(heart failure,HF)的发生发展密切相关。同期研究发现[2],参麦注射液(SMI)有提高血清EDLS浓度的重要作用,并呈现一定的量效、时效关系,该实验为进一步了解SMI对心衰犬组织EDLS浓度的影响。

    1 材料与方法

    1.1 实验动物 健康成年(12个月龄以上)杂种犬6只,体重(11.7±0.6)kg,雌雄不拘,饲养于天津中医药大学实验动物中心,适应性饲养1周以上备用。

    1.2 实验仪器与药品 MP-150型16道生理记录仪(美国BIOPAC),SC-3型电动呼吸机(上海医疗设备厂),LOGIQ-400数字彩色多普勒超声心动仪(美国GE公司),MH-01型自动放射免疫伽玛计数仪(北京核仪器厂)。SMI(杭州正大青春宝药业生产,批号:0508293)。

    1.3 心衰犬模型复制 采用结扎犬冠状动脉前降支法复制心衰动物模型[3]。

    1.4 模型鉴定 结扎冠脉后可见梗死区域颜色变暗,心肌呈现暗红色,心室前壁运动减弱,心外膜心电图ST段明显抬高。结扎冠状动脉术后(1~4)周,实验犬逐渐出现倦怠、活动减少、反应迟缓、呼吸增快、食欲下降、稀便等征象 ......
