EGF-CP-HA水凝胶对大鼠皮肤创面愈合的影响(1)

     [摘要] 目的 通過胶原蛋白肽-透明质酸负载表皮生长因子(EGF-CP-HA)水凝胶在大鼠全层皮肤缺损伤口的应用来探讨其对皮肤伤口愈合的影响。方法 90只SD大鼠,麻醉后制备伤口创面动物模型,随机分为生理盐水(NS)组、透明质酸(HA)水凝胶组、胶原蛋白肽-透明质酸(CP-HA)水凝胶组、透明质酸负载表皮生长因子(EGF-HA)水凝胶组和EGF-CP-HA水凝胶组,每组18只。后4组大鼠在其皮肤伤口处分别贴敷相应水凝胶治疗。观察并记录创面愈合情况。分别在相应时间点麻醉大鼠后取伤口及其周缘1 cm的组织,行苏木精-伊红、Masson、免疫组化染色来观察损伤区域不同时间的组织病理变化、胶原纤维分布、血管内皮生长因子-α(VEGF-α)和转化生长因子-β1(TGF-β1)的表达。结果 EGF-CP-HA水凝胶组大鼠皮肤伤口愈合时间最短(F=6.46,P<0.01)。苏木精-伊红染色显示,EGF-CP-HA水凝胶组创面炎性反应较NS组轻,毛细血管及胶原纤维生成较其他组多;Masson染色显示,EGF-CP-HA水凝胶组胶原纤维较其他组多,排列规则、有序,更接近正常皮肤;免疫组化染色显示,EGF-CP-HA水凝胶组TGF-β1和VEGF-α的表达强度均显著高于NS组(F=12.51~18.64,P<0.05)。结论EGF-CP-HA水凝胶可减轻皮肤创面的炎症反应,提高VEGF-α和TGF-β1的表达水平,明显促进伤口愈合。

    [ABSTRACT] Objective To explore the effect of collagen peptide-hyaluronic acid loaded epidermal growth factor (EGF-CP-HA) hydrogel on skin wound healing in rats with full-thickness skin defect. Methods Ninety Sprague-Dawley rats were randomly divided into saline (NS) group, hyaluronic acid (HA) hydrogel group, collagen peptide-hyaluronic acid (CP-HA) hydrogel, hyaluronic acid-loaded epidermal growth factor hydrogel (EGF-HA) group, and EGF-CP-HA hydrogel group, with 18 rats in each group. The rats in the latter four groups were treated with corresponding hydrogels on their skin wounds. Wound healing was observed and recorded. After the rats were anesthetized at the corresponding time point, tissue samples at the wound and 1 cm from the wound were taken and subjected to hematoxylin-eosin, Masson, and immunohistochemical staining to observe the histopathological changes, the distribution of collagen fibers, and the expression of vascular endothelial growth factor-α (VEGF-α) and transforming growth factor-β1 (TGF-β1) in the injured area at different time points. Results The wound healing time of rats in EGF-CP-HA hydrogel group was the shortest (F=6.46,P<0.01). Hematoxylin-eosin staining showed that the EGF-CP-HA hydrogel group had milder inflammatory reaction of wound surface than the NS group, and had more formation of capillaries and collagen fibers than other groups; Masson staining showed that collagen fibers were more regularly arranged and closer to normal skin in the EGF-CP-HA hydrogel group than in other groups. Immunohistochemical staining showed that the expression of TGF-β1 and VEGF-α was significantly higher in the EGF-CP-HA hydrogel group than in the NS group (F=12.51-18.64,P<0.05). Conclusion EGF-CP-HA hydrogel can alleviate the inflammatory reaction of skin wounds, increase the expression levels of VEGF-α and TGF-β1, and significantly promote wound healing., http://www.100md.com(程连强 施春英 郭恩慧 邱鲁阳 夏玉军)
