壳寡糖对四氯化碳诱导的肝损伤的保护作用

     摘要: 目的研究壳寡糖(COS)对四氯化碳(CCl4)诱导的肝损伤的保护作用。方法雄性昆明种小鼠24只分为3组,正常对照组及CCl4模型对照组予以相应体积的生理盐水,COS组灌胃给予COS(1.5 g/kg),后2组于第12天腹腔注射CCl4(20 mg/kg)形成肝损伤模型。测定3组小鼠血清中丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)活性,以及肝匀浆中丙二醛(MDA)含量和DNA损伤情况。结果COS组与CCl4模型对照组相比,ALT,AST活性和MDA含量分别低62.2%,52.9%和34.3%。DNA电泳结果示,COS组与CCl4模型对照组的DNA链都形成一系列约1 000 bp的DNA片断。结论COS对CCl4诱导的肝损伤有较明显的保护作用,但是不能减轻DNA的氧化性损伤。

    Protective Effect of Chitosan Oligosaccharide against Liver Damage Induced by Carbon Tetrachloride

    Abstract:Objective To study the protective effect of chitosan oligosaccharide (COS) against liver damage induced by carbon tetrachloride (CCl4). Methods Twenty four male ICR mice were divided into three groups of normal control, CCl4 control and COS pretreated groups. The mice in COS pretreated group were pretreated with COS (1.5g/kg weight) once daily for 12 consecutive days, while the mice in normal control and CCl4 control groups were pretreated with correspondingly volumetric saline also for 12 days. Then, the mice in CCl4 control and COS

    pretreated groups were intraperitoneally injected with CCl4 (20mg/kg weight) to make liver damage models. Blood was collected and serum was separated for the determination of activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST).
