     [摘要] 目的:探讨非小细胞肺癌患者外周血中CD4+CD25+调节性T细胞(regulatory T cell, Treg)数量变化及其相关基因Foxp3 mRNA的表达改变,探讨CD4+CD25+调节性T细胞在诱导肿瘤免疫耐受中的作用。方法:采用流式细胞术检测患者外周血中CD4+CD25+调节性T细胞数量变化;采用半定量RT-PCR方法检测肺癌患者外周血PBMC中Foxp3 mRNA的表达水平。选取30名非肿瘤患者作为对照。结果:肺癌患者外周血中CD4+CD25+调节性T细胞数量明显高于正常组(P<0.05);Foxp3 mRNA在肺鳞癌和肺腺癌患者外周血PBMC中的表达明显增强(P<0.05)。结论:CD4+CD25+调节性T细胞可能在肺癌免疫耐受中发挥重要作用。

    The changes of CD4+CD25+ T cells and expression of Foxp3 mRNA in peripheral blood mononuclear cells from patients with non-small-cell lung carcinoma

    [Abstract] Objective: To investigate the changes of CD4+CD25+ T cells and expression of Foxp3 mRNA in peripheral blood mononuclear cells from patients with non-small-cell lung carcinoma. Methods: Flow cytometry was used to analyze the contents of CD4+CD25+ regulatory T cells in peripheral blood from the patients with non-small-cell lung carcinoma and patients without cancer as control. The expression of Foxp3 mRNA in peripheral blood mononuclear cells was detected by RT-PCR. Results: The population of CD4+CD25+ regulatory T cells in peripheral blood from the patients with non-small-cell lung carcinoma was significantly higher in comparison with that of control (P<0.05). The expression of Foxp3 mRNA in patients with lung carcinoma was also significantly higher in comparison with that of control (P<0.05). Conclusion: CD4+CD25+ regulatory T cells and its related Foxp3 may play an important role in the immune of tumor.

    肺癌是当今世界对人类健康危害最大的肿瘤之一,且其发病率有不断上升的趋势。预计到2025年,每年将有90万人死于肺癌,中国将成为肺癌第一大国。因此,对肺癌的研究意义重大。

    CD4+CD25+调节性T细胞(regulatory T cell, Treg)是健康个体T细胞库的组成部分,占CD4+细胞的5%~10%。该细胞群具有抑制免疫效应的作用,能特异性抑制T细胞对外源和自体抗原的免疫反应,因此在维持对自身成分免疫耐受的同时,也可能阻止机体对自体同源肿瘤细胞的免疫。目前普遍认为,CD4+CD25+Treg很可能影响免疫系统对肿瘤抗原的应答,从而影响肿瘤的发生和发展。Foxp3是特异性地表达于CD4+CD25+Treg表面的一种转录因子,可作为该类细胞的特异标志,并调节其发育和功能。

    本文通过检测非小细胞肺癌患者外周血中CD4+CD25+Treg的数量变化及其相关基因Foxp3 mRNA的表达,探讨CD4+CD25+Treg与非小细胞肺癌发生、发展的关系。
