稳定大鼠原位肝移植模型的建立(1)

     [摘要] 目的:建立稳定大鼠原位肝移植模型,探讨其方法与技术。方法:用改进的二袖套法行140例SD→LEW大鼠原位肝移植,术中门静脉、肝下下腔静脉用袖套法吻合,肝上下腔静脉用缝合法吻合,胆管内支架法完成胆道重建。结果:供体手术时间为(27.0±3.1) min;供肝修整时间为(11.0±2.1) min;受体手术时间为(30.0±2.5) min;无肝期为(15.0±2.6) min。手术成功率(术后存活2 d以上)为87%,1周存活率为81%,1个月存活率为72%。结论:提高手术技巧,熟悉各种并发症的原因和预防处理措施,可以减少并发症的发生,提高手术成功率,延长大鼠术后存活时间。

    The establishment the rat stable model of orthotopic liver transplantation

    [Abstract] Objective: To establish the rat stable model of orthotopic liver transplantation, and explore the methods and technology. Methods: 140 cases of SD→LEW rat liver transplantation had been done with improved two-cuff method in the portal vein, hepatic inferior vena cava under the law in line with cuff, liver superior and inferior vena cava was sutured with suture line. Biliary tract reconstruction was completed in bile duct stent law. Results: Donor surgery time was (27.0±3.1) min; liver repaired time was (11.0±2.1) min; surgery receptor time was (30.0±2.5) min; no liver time was (15.0±2.6) min. The success rate (survived more than 2 days after operation) was 87%. One week survival rate was 81%, one month survival rate was 72%. Conclusion: Enhancing the surgery skill and understanding the reasons and prevention and treatments of each kinds of complication may reduce the occurrence of complications, raise the surgery success rate and lengthen the survival time of rats.

    建立大鼠肝移植模型是缺血-再灌注损伤、排斥反应以及免疫耐受研究的有效手段。本实验采用Kamada“二袖套法”[1],结合一些相关报道[2-3]进行相应的改进,成功地建立了稳定大鼠肝移植模型,为后期的移植免疫研究奠定了基础。

    1 材料与方法

    1.1 实验动物

    供体SD大鼠与受体LEW大鼠各70只,体重220~300 g,由山西医科大学实验动物中心提供。供、受体大鼠体重基本相同[4]。1.2 套管的制备
