中药启膈散免疫调节作用的体外实验研究(1)

     [摘要] 目的:探讨启膈散抗肿瘤作用的机制,为启膈散治疗肿瘤提供客观依据。方法:将实验细胞分为启膈散组、刀豆蛋白A(concanavalin A,ConA)组、脂多糖(lipopolysaccharide,LPS)组和空白对照组,ConA组及LPS组分别给予ConA、LPS 5 μg/ml,空白对照组不给药,采用MTT法观察50.00、25.00、12.50、6.25、3.13、1.63、0.78 mg/ml启膈散水提取物对小鼠脾细胞体外增殖的促进作用,50.00、5.00、0.50 mg/ml时对小鼠脾细胞分泌IL-2的影响;50.00、5.00、0.50 mg/ml时对小鼠单核细胞分泌TNF-α的影响;50.00、5.00、0.50 mg/ml时对NK细胞肿瘤细胞杀伤活性的影响。结果:启膈散显著刺激小鼠脾细胞增殖,且存在明显的量效关系;能显著提高小鼠脾细胞分泌IL-2的活性;促进小鼠单核细胞分泌TNF-α作用显著;可提高NK细胞对靶细胞的杀伤率。结论:启膈散的抗肿瘤活性可能与其免疫调节功能有关。

    In vitro experiment of TCM Qige Powder on immune regulation

    [Abstract] Objective: To explore the anti-tumor mechanism of Qige Powder, and provide objective basis for treating tumor by Qige Powder. Methods: The experimental cells were divided into Qige Powder group, concanavalin A (ConA) group, lipopolysaccharide (LPS) group and control group, ConA group and LPS group were given 5 μg/ml ConA and LPS respectively, the control group was not given drug. The promoting effect of Qige Powder water extract with doses of 50.00, 25.00, 12.50, 6.25, 3.13, 1.63 and 0.78 mg/ml on in vitro mice spleen cells was detected with MTT assay; the effect of Qige Powder water extract with doses of 50.00, 5.00 and 0.50 mg/ml on IL-2 secreted by mice spleen cells, the effect of Qige Powder water extract with doses of 50.00, 5.00 and 0.50 mg/ml on TNF-α secreted by mice monocytes and the cytotoxic activity of Qige Powder water extract with doses of 50.00, 5.00 and 0.50 mg/ml on tumor cells caused by NK cells were observed respectively. Results: Qige Powder could significantly promote the increase of mice spleen cells, and it should an obvious dose-dependent relationship; it could also improve the IL-2 level secreted by mice spleen cells and the secretion of TNF-α by mice monocytes, and enhanced the killing rate of NK cells to target cells ......
