人参瓜蒌莪术汤对小鼠Lewis肺癌端粒酶活性的影响(1)

     [摘要] 目的:研究人参瓜蒌莪术汤(RGE)对Lewis肺癌模型小鼠的治疗作用及机制。方法:建立小鼠Lewis肺癌移植瘤模型,分为模型对照组(NS组),环磷酰胺对照组(CP组),RGE高、中、低剂量组。在计算抑瘤率的基础上,运用PCR-ELISA法检测瘤组织端粒酶活性。结果:RGE低、中、高剂量组均能明显降低小鼠Lewis肺癌瘤重(P<0.01),中剂量组抑瘤率达47.88%;RGE低、中、高剂量组均能明显降低瘤组织细胞端粒酶的活性(P<0.01),中剂量组对端粒酶活性的抑制作用显著强于CP组(P<0.01)。结论:RGE具有抑制小鼠Lewis肺癌生长的作用,其作用机制可能与降低肿瘤组织端粒酶活性有关。

    Effect of Renshengualouezhu Decoction on telomerase activity of Lewis lung cancer in mice

    [Abstract] Objective: To explore the influence of treatment and mechanism of Renshengualouezhu Decoction (RGE) on Lewis lung cancer in mice. Methods: Mice Lewis lung cancer models were established, and divided into 5 groups: model control group (NS group), low-dose RGE group, middle-dose RGE group, high-dose RGE group and Cyclophosphamide group(CP group). Then calculated the inhibition rate and used PCR-ELISA to detect the activity of telomerase. Results: The tumor weight of low-dose RGE group, middle-dose RGE group, high-dose RGE group and CP group were significantly decreased (P<0.01). Inhibition rate in the middle-dose RGE group reached 47.88%. RGE could reduce the telomerase activity on all dose levels of low, middle or high (P<0.01). The middle-dose RGE group was significantly better than the CP group (P<0.01). Conclusion: RGE can significantly inhibit the growth of mice Lewis lung cancer. It may be related to its ability to reduce telomerase activity.

    肺癌是严重危害人类健康的恶性肿瘤之一,也是全世界目前发病率和死亡率最高的癌症。中医药疗法治疗肺癌在稳定病灶、延长生存时间、改善生存质量、减轻放、化疗的毒副作用和增强放、化疗的效果等方面显示出独特的疗效与作用优势[1-3]。人参瓜蒌莪术汤(RGE)是全国知名老中医邱幸凡教授40年临床治疗肺癌的经验方,也是其“补肺通络解毒法”治疗肺癌的代表方剂,主要具有补肺、通络、解毒、消瘤的功效,临床疗效显著。本实验选用小鼠Lewis肺癌模型,在观察RGE抑瘤率的基础上,运用PCR-ELISA法检测肿瘤组织端粒酶活性,初步探讨该复方的作用效果与机制,为其临床应用提供实验依据。

    1 材料与方法

    1.1 材料

    1.1.1 实验动物及瘤株健康C57BL/6J小鼠50只,雌性,鼠龄6~8周,体重(20±2) g,由江苏省实验动物中心提供;Lewis肺癌细胞株(Lewis lung cancer,3LL),由南京市凯基生物制品有限公司提供。
