HPLC法测定十滴水中五种活性成分的含量(1)

     [摘要] 目的:建立HPLC法同时测定十滴水中的芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚含量的方法。方法:色谱柱为Shimadzu VP-ODS柱(4.6 mm×250 mm, 5 μm),流动相为乙腈-0.1%磷酸梯度洗脱,流速为1.0 ml/min,检测波长为254 nm,柱温30℃。结果:5种成分分别在0.042~0.840 μg(r=0.999 6)、0.168~3.352 μg(r=0.999 8)、0.084~1.680 μg(r=0.999 7)、0.093~1.852 μg(r=0.999 9)和0.174~3.490 μg(r=0.999 4)内呈良好线性关系,平均回收率分别为99.60% (RSD=0.20%)、99.56% (RSD=0.36%)、98.95% (RSD=0.55%)、98.76% (RSD=0.97%)和98.79% (RSD=1.08%)。结论:本法简便、快速、准确,可用于十滴水的质量控制。

    Determination of five effective constituents in Shidishui by HPLC

    [Abstract] Objective: To establish a method for the simultaneous determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in Shidishui by HPLC. Methods: The sample was separated on a Shimadzu VP-ODS (4.6 mm×250 mm, 5.0 μm) with acetonitrile-0.1% phosphoric acid in gradient elution. The flow rate was 1.0 ml/min, the detection wavelength was 254 nm and the column temperature was 30℃. Results: Aloe-emodin, rhein, emodin, chrysophanol and physcion had good linear relationship in the range of 0.042-0.840 μg (r=0.999 6), 0.168-3.352 μg (r=0.999 8), 0.084-1.680 μg (r=0.999 7), 0.093-1.852 μg (r=0.999 9) and 0.174-3.490 μg (r=0.999 4). Their average recoveries were 99.60% (RSD=0.20%), 99.56% (RSD=0.36%), 98.95% (RSD=0.55%), 98.76% (RSD=0.95%), and 98.79% (RSD=1.08%), respectively. Conclusion: The method is rapid, simple, accurate, and applicable for the quality control of Shidishui.

    十滴水为常用中成药,是《中国药典》2005年版一部收载品种,主要由大黄、樟脑、干姜等7味中药组成,具有健胃,祛暑的功效。主要用于由中暑引起的头昏、恶心、腹痛、肠胃不适[1]。方中大黄性苦、寒,具有泻热通肠、凉血解毒的作用,在方中起主要疗效,蒽醌类衍生物是大黄的主要活性成分[2-7]。目前,对于十滴水中单体的含量测定有少量的文献报道[7-9],但同时测定多个成分的含量未见文献报道。本研究采用HPLC法同时测定大黄中5种活性成分芦荟大黄素(1)、大黄酸(2)、大黄素(3)、大黄酚(4)和大黄素甲醚(5)的含量,为该制剂的质量控制和临床应用提供参考。
