参蛇偏瘫胶囊联合尼莫地平预处理对脑缺血后再灌注神经细胞凋亡的影响(1)

     [摘要] 目的:探讨参蛇偏瘫胶囊联合尼莫地平预处理对脑缺血后再灌注神经细胞凋亡的影响。方法:采用夹闭小鼠双侧颈总动脉造成脑缺血模型。将50只小鼠随机分成五组,即假手术组、模型组、尼莫地平组、参蛇偏瘫胶囊组、参蛇偏瘫胶囊联合尼莫地平预处理组(联合组),每组各10只。尼莫地平组、参蛇偏瘫胶囊组、联合组三组缺血前30 min分别静脉给予尼莫地平0.2 mg/kg、腹腔注射参蛇偏瘫胶囊10 ml/kg以及二者同时联合处理,缺血5 min后分别再灌注72 h。取脑片观察各组脑细胞凋亡的变化情况,并采用免疫组化染色评定Bcl-2蛋白反应强度。结果:联合组的凋亡细胞百分数明显低于其他各组(P<0.05),而Bcl-2蛋白免疫反应强度高于其他各组(P<0.05)。结论:脑缺血前经参蛇偏瘫胶囊联合尼莫地平预处理后能明显减少细胞凋亡的发生,且优于单独使用二者,此与Bcl-2蛋白表达增强有关。

    Influences of preconditioning with Shenshe Piantan Capusle and Nicardipine on neuron apoptosis following acute cerebral ischemia-reperfusion

    [Abstract] Objective: To study the influences of preconditioning with Shenshe Piantan Capusle and Nicardipine on neuron apoptosis following acute cerebral ischemia-reperfusion. Methods: To make occlusion of cephalic artery in mice as model of cerebral ischemia. 50 mice were randomly divided into 5 groups, sham operative control group, model group, Nimodipine group, Shenshe Piantan Capusle group (SPC group) and Shenshe Piantan Capusle combined with Nimodipine group (combination group), each 10 mice. After 5 min cerebral ischemia and following 72 h reperfusion. Nimodipine group, Nimodipine (0.2 mg/kg) was iv 30 min before ischemia; SPC group, SPC (10 ml/kg) was injected into abdominal cavity 30 min before ischemia; combination group, SPC (10 ml/kg, injected into abdominal cavity) associated Nimodipine (0.2 mg/kg, iv) 30 min before ischemia. Neuron apoptosis was identified by TUNEL and Paraffin sections of cortex were used for Bcl-2 protein immunohistochemical staining. Results: The percentage of apoptotic cells in combination group was significantly lower than those of other groups (P<0.05) and Bcl-2 protein immunoreactivity significantly increased than those of the other groups (P<0.05).
