夹带剂对超临界CO2萃取丹参中丹参酮ⅡA的影响(1)

     [摘要] 目的:对夹带剂对超临界CO2萃取丹参中丹参酮ⅡA的影响进行研究。方法:从夹带剂总量和单次加入夹带剂量两个方面分析,设计以丹参酮ⅡA含量为指标的单因素实验。结果:在萃取压力35 MPa、萃取温度65℃、解析压力6 MPa、解析温度45℃的条件下,提取时间3 h,95%乙醇作为夹带剂,夹带剂用量是:药材量的200%(质量分数);单次加入夹带剂量是:每隔0.5 h加入一次,分别是药材量的0.5倍,0.4倍,0.4倍,0.4倍,0.2倍和0.1倍。丹参酮ⅡA提取率均在90%以上,药渣中丹参酮ⅡA残留率均在6%左右。结论:夹带剂加入总量和单次加入量均对萃取结果有显著影响。

    Effects of entrainer in extracting of tanshinone ⅡA from the root of Salvia miltiorrhiza Bge. by supercritical carbon dioxide fluid

    [Abstract] Objective: To study the effects of entrainer in extracting of tanshinone Ⅱ A from the root of Salvia miltiorrhiza Bge. by supercritical carbon dioxide fluid. Methods: The total dosage of entrainer and a single dose were analyzed and a single-factor experiment was designed with the content of tanshinone ⅡA as the index. Results: When the extraction was 35 MPa,extraction temperatrue was 65℃, separate pressure was 6 MPa, and separate temperature was 45℃, extraction time was 3 h and entrainer was ethanol(95%), the total dosage of entrainer was 200%;the single dose of entrainer was Chinese raw material for medicine 0.5 times, 0.4 times, 0.4 times, 0.4 times, 0.2 times and 0.1 time. Tanshinone ⅡA extraction rate was above 90%, in dregs of medical decoction remain rate was about 6%. Conclusion:The total dosage and the single dosage of the added entrainer have the remarkable influence to the extract result.

    丹参为唇形科植物丹参Salvia miltiorrhiza Bge.的干燥根及根茎。丹参酮ⅡA是中药丹参的主要有效成分,具有保护心肌缺血缺氧、抑制血小板聚集的功能,还具有明显的抗肿瘤作用[1-2]。近年来,超临界CO2萃取被应用于丹参中丹参酮ⅡA的萃取[3-4],总结分析,夹带剂均采用无水乙醇和95%乙醇,但是其用量却因各工艺不同有较大差别,质量分数从30%到200%不等[5-8]。因此,本文重点考察夹带剂的使用方法对萃取结果的影响,进而对超临界CO2萃取丹参中丹参酮ⅡA工艺进行研究。
