[摘要] 目的：建立反相高效液相色谱法同时测定消炎片中绿原酸和秦皮乙素的含量。方法：采用Agilent Eclipse XDB-C18(250 mm×4.6 mm，5 μm)色谱柱，以乙腈-0.04%磷酸溶液为流动相，梯度洗脱；检测波长为328 nm；流速1.0 ml/min；进样量10 μl；柱温35℃。结果：绿原酸和秦皮乙素的线性范围分别为2.515~30.18 μg/ml(r=0.999 9，n=5)和6.388~76.65 μg/ml (r=1.000 0，n=5)；提取回收率分别为98.99%(RSD为0.2%，n=6)和98.93%(RSD为0.5%，n=6)。结论：本法操作简便、快速、结果准确，可用于消炎片的质量控制。

Simultaneous determination of chlorogenic acid and esculetin in Xiaoyan Tablets by HPLC

[Abstract] Objective: To establish RP-HPLC method for simultaneous determination of chlorogenic acid and esculetin in Xiaoyan Tablets. Methods: Column packed with Agilent Eclipse XDB-C18 (250 mm×4.6 mm , 5 μm) was used; the mobile phase was acetonitrile-0.04%H3PO4 by gradient elution and the detection wavelength was 328 nm; the flow rate was 1.0 ml/min, the injection volume was 10 μl, the column temperature was 35℃. Results: The linear response of chlorogenic acid ranged from 2.515 to 30.18 μg/ml, r=0.999 9 (n=5); the linear response of esculetin ranged from 6.388 to 76.65 μg/ml, r=1.000 0 (n=5); the average recovery of chlorogenic acid was 98.99%, with RSD of 0.2% (n=6). The average recovery of esculetin was 98.93% with RSD of 0.5%(n=6). Conclusion: The method is convenient, quick and accurate, and can be used for the quality control of Xiaoyan Tablets.

消炎片由蒲公英、紫花地丁、野菊花、黄芩4味中药加工、提取制成，具有抗菌消炎之功效[1]。现行质量标准[1]无含量测定方法。目前有多篇文献[2-4]关于HPLC法测定消炎片中绿原酸、秦皮乙素含量的报道，未见采用HPLC法同时测定两组分的报道，本文建立了同时测定消炎片中野菊花的有效成分绿原酸和紫花地丁有效成分秦皮乙素的含量测定方法，本方法简便、准确、重现性好，可用于消炎片的质量控制。
1 仪器与试药
1.1仪器
HP1100高效液相色谱仪，HP1100工作站(自动进样，紫外检测器)，Sartorius电子天平(CP225D)，KQ-400KDE型高功率数控超声波清洗器，TU-1901双光束紫外-可见分光光度计(北京普析通用仪器有限责任公司)。
