血红素加氧酶1在左心室肥厚和心力衰竭中的表达及其意义(1)

     [摘要] 目的:探讨在左心室肥厚向心力衰竭的转变过程中心肌细胞凋亡和血红素加氧酶1的表达及意义。方法:采用缩窄腹主动脉造成心力衰竭模型,术后分为3组,假手术组10只、心肌肥厚组10只和心力衰竭组10只。假手术组、心肌肥厚组于6周时处死,心力衰竭组于14周时处死,处死前行血流动力学、心肌肥厚检测,TUNEL法检测心肌细胞凋亡,Western Blot法检测心肌Caspase-3和血红素加氧酶1的表达。结果:心力衰竭组较心肌肥厚组、假手术组心肌细胞凋亡增加(P<0.05),Caspase-3表达增加(P<0.01);心肌肥厚组较心力衰竭组、假手术组血红素加氧酶1表达增加(P<0.05)。结论:心肌细胞凋亡在心脏由肥厚向衰竭的转变过程中起重要的作用,与血红素加氧酶1的改变有关。

    Expression and significance of heme oxygenase 1 in cardiomyocyte from left ventricular hypertrophy to heart failure

    [Abstract] Objective: To observe the role and significant of apoptosis and heme oxygenase 1 in the transition from Left ventricular hypertrophy (LVH) to heart failure (HF). Methods: Pathological models of heart failure were established by constriction of abdominal aorta of rats partly. After operation, 30 rats were divided randomly into 3 groups, sham-operated group (n=10), ventricular hypertrophy group (n=10), heart failure group (n=10). After 6 weeks, the rats of sham-operated group and ventricular hypertrophy group were killed. After 14 weeks, the rats of heart failure group were killed. The hemodynamic parameters and ventricular hypertrophy were studied. Apoptosis were stained in situ by using TUNEL. The monitoring of heme oxygenase 1 and Caspase-3 protein of cardiomyocyte was determined by western blot. Results: Compared with sham-operated group and ventricular hypertrophy group, cardiomyocyte apoptotic events and expression of Capase-3 were increased in heart failure group (P<0.05). Expression of the heme oxygenase 1 was increased in the ventricular hypertrophy group (P<0.01), whereas that of heme oxygenase 1 was decreased in heart failure group compared with ventricular hypertrophy group (P<0.05).
