干扰STAT3的siRNA真核表达载体对人卵巢癌的侵袭转移性的相关研究(1)

     [摘要] 目的:通过构建针对STAT3的siRNA真核表达载体,研究STAT3对卵巢癌SKOV3细胞侵袭转移能力的影响。方法:使用软件Primer Premier 5设计siRNA靶序列,在合成修饰靶序列之后,构建siRNA-STAT3-脂质体复合物,并转染卵巢癌SKOV3细胞;通过Western blot检测细胞STAT3的表达,通过Transwell小室及对卵巢癌SKOV3细胞侵袭相关蛋白MMP-2的检测,探讨siRNA-STAT3对卵巢癌细胞侵袭力的影响。结果:构建干扰STAT3的siRNA真核表达载体转染SKOV3细胞可显著抑制细胞中STAT3的蛋白表达;与SKOV3细胞对照组和SKOV3细胞阴性对照组相比,SKOV3细胞siRNA-STAT3组中细胞侵袭转移能力明显下降。结论:构建干扰STAT3的siRNA真核表达载体可有效抑制STAT3的表达及卵巢癌SKOV3细胞的侵袭转移能力。

    The study on relationship between STAT3 eukaryotic expression vector of small interference RNA and invasion and metastasis of ovarian cancer

    [Abstract] Objective: To construct small interference RNA (siRNA) expression vector of the signal transducer and activator of transcription 3 (STAT3), and take a research on the form and transfer ability of ovarian cancer cells (SKOV3). Methods: STAT3 specific oligonucleotides were designed by software Primer Premier 5, and siRNA-STAT3-LipofectamineTM2000 complex was constructed after combining and embellishing the target sequences, a transfection was made in ovarian cancer cells (SKOV3), the expression of STAT3 was detected by Western blot way, transwell matrigel invasion assays was used to test the invasive power, and the protein expression level of MMP-2 was examined by Western blot analysis, transfer inlfluence of the ovarian cancer cells was explored by siRNA-STAT3. Results: The protein expression levels of STAT3 in ovarian cancer cells (SKOV3) were decreased with transfection of siRNA-STAT3, and in the siRNA-STAT3 transfection group, the invasion and proliferation ability ofovarian cancer cells were greatly suppressed which group compared with SKOV3 control group and SKOV3 negative control group ......
