复方血竭微乳对豚鼠皮肤刺激性实验观察(1)

[摘要] 目的：对复方血竭微乳进行皮肤刺激性试验，以了解其安全性。方法：将复方血竭微乳及空白微乳约0.5 ml涂于豚鼠完整皮肤及破损皮肤，在完整皮肤和破损皮肤进行一次给药和多次给药皮肤刺激性试验，观察复方血竭微乳及其空白微乳是否会引起豚鼠皮肤刺激性。结果：复方血竭微乳及其空白微乳一次给药对完整皮肤组和破损皮肤组均未出现红斑、水肿等现象，无刺激现象；复方血竭微乳及其空白微乳多次给药对破损皮肤有轻微刺激作用。结论：复方血竭微乳多次给药对豚鼠完整皮肤无刺激性，对破损皮肤有轻度刺激性，但给药后72 h刺激性反应自行消退。

Observation of skin irritation experiment in guinea pigs with Compound Resina Draconis Microemulsion

[Abstract] Objective: To study the safety of Compound Resina Draconis Microemulsion (CRDM) in skin irritation experiments in order to know its safety. Methods: 0.5 ml of CRDM and its blank Microemulsion were spreaded on the normal or damaged skin of guinea pigs once or many times to observe whether CRDM and its blank Microemulsion could cause skin irritation in guinea pigs. Results: CRDM and its blank Microemulsion didn't cause erythema, dropsy and other irritation conditions on the normal or damaged skin after administration once. CRDM and its blank Microemulsion caused light irritation reaction on the damaged skin after multiple administrations. Conclusion: CRDM causes no irritation on the normal skin and light irritation on the damaged skin of guinea pigs after multiple administrations, but the irritation reaction disappears in 72 hours after administration spontaneously.

中药治疗烧伤及其他化学物质引起的皮肤损伤具有多成分、多效应的综合优势。由血竭、川芎、肉桂等中药组成的经验方具有去腐生肌、促进局部血液循环和抑菌等作用[1-4]，使多效应机制促进损伤皮肤的快速愈合。为了进一步提高疗效和便于临床使用，在保持疗效的基础上，利用微乳具有表面张力较低、易于润湿皮肤、组织透过性好的优点[5]，将该经验方提取活性成分后制备成O/W型微乳。为了观察其在局部给药后对皮肤的刺激性，笔者采用豚鼠进行了皮肤毒性试验，现将结果报道如下：
1 材料与方法
1.1 仪器与试剂
十万分之一电子天平(梅特勒)；复方血竭微乳及空白微乳(第四军医大学唐都医院药剂科提供，批号：20100910)；生理盐水(西安京西双鹤药业有限公司，批号：101209121)；75%乙醇消毒液(德州安捷高科消毒制品有限公司，批号：20100712)。
1.2 动物
白色豚鼠：体重300~400 g，清洁级，雌雄各半[购于第四军医大学动物中心，合格证号：SCXK(军)字第2002-005号]，实验前在动物房适应7 d。动物房温度：(25±1)℃，相对湿度：50%~70%。
1.3 实验方法
1.3.1 实验前豚鼠皮肤处理
