NKX2.5 基因修饰骨髓间充质干细胞对心肌梗死大鼠心功能的影响(1)

     [摘要] 目的:以NKX2.5融合绿色荧光蛋白基因(NKX2.5-pEGFP)质粒转染大鼠骨髓间充质干细胞(MSCs),将基因修饰后MSCs移植入大鼠心肌梗死模型中,评价其对心功能的影响。方法:体外分离培养大鼠MSCs,构建NKX2.5-pEGFP基因质粒,以lipo2000将质粒转染入MSCs。将基因修饰后MSCs植入心肌梗死后10 d的大鼠心肌梗死区(MSCs组,n=20),同时将注射等体积无血清改良依格尔(DMEM)培养液于心肌梗死区的大鼠作为DMEM组(n=20)。移植注射后4周,通过检测左心室射血分数(LEVF)、左室缩短分数(FS)等研究其对心功能的影响。结果:移植4周后,MSCs组大鼠LEVF[(79±4)%]较DMEM组[(47±3)%]显著提高(P<0.01), MSCs组大鼠FS[(48±9)%]较DMEM组[(25±6)%]显著提高(P<0.01)。结论:移植NKX2.5基因修饰后的MSCs能够改善心肌梗死大鼠心功能。

    Heart function effect of mesenchymal stem cells modified by NKX2.5 in infracted myocardium in rats

    [Abstract] Objective: To transfect NKX2.5-pEGFP plasmid into mesenchymal stem cells (MSCs) and transplant the gene modified MSCs in rat myocardial infarction model to estimate the effect on heart function. Methods: MSCs were isolated culture in vitro. NKX2.5-pEGFP plasmid was constructed and transfected into MSCs. The gene modified MSCs was transplanted in the area of rat myocardial infarction model after 10 days (MSCs group, n=20). The equal volume of none blood serum DMEM was injected in myocardial infarction model as DMEM group (n=20). The effect on heart function was estimated by detecting left ventricular ejection fraction (LVEF) and fractional shortening (FS) after injecting 4 weeks. Results: 4 weeks after transplantation, the LEVF value of MSCs group [(79±4)%] increased significantly compared to that of DMEM group [(47±3)%] (P<0.01) and the FS value of MSCs group [(48±9)%] increased significantly compared to that of DMEM group [(25±6)%] (P<0 ......
