应用Fluoro-Jade C评价小鼠匹罗卡品模型中纹状体神经变性(1)

     [摘要] 目的:应用Fluoro-Jade C(FJC)染色方法在小鼠匹罗卡品癫痫模型中探测纹状体结构中神经元的变性情况,以了解纹状体结构在慢性颞叶癫痫发生中的病理神经基础。方法:雄性昆明小鼠10只(对照组5只,模型组5只)。在癫痫持续状态后3 d处死模型组小鼠。在纹状体水平切制冠状切片,行FJC染色,在荧光显微镜下观察FJC阳性细胞的形态和在纹状体(尾状壳核)中的整体分布情况。结果:在模型组,FJC染色的脑切片上清楚显示FJC阳性变性细胞呈亮黄绿色,呈神经元形态,胞体和突起均清晰显示。在纹状体(尾状壳核)内背侧1/3和尾侧部分出现许多FJC阳性细胞。结论:本研究运用FJC染色技术在小鼠匹罗卡品癫痫模型中显示纹状体中发生了神经元变性,提示纹状体可能在慢性颞叶癫痫的发生机制中起重要作用。

    Fluoro-Jade C staining in the assessment of neurodegeneration in the striatum of Pilocarpine-treated mice

    [Abstract] Objective: To detect degenerative neurons by Fluoro-Jade C (FJC) that may occurred in the striatum of pilocarpine-treated mice and to understand the pathologically neural basis of striatum in chronic epilepsy. Methods: Male adult mice (Kunming stain, n=10) were divided into Pilocarpine-treated group (n=5) and control group (n=5). Pilocarpine-treated mice were sacrificed at 3 d after status epilepticus. Brain coronal sections in the striatum levels were subject to FJC staining. Finally, FJC-stained brain and overall distribution of the Fluoro-Jade C positive cells sections were examined under an epifluorescence microscope to identify the shape in the striatum (caudate putamen). Results: The FJC-positive staining cells showed a bright green color in the somas and fine processes with neuronal profiles. Numerous degenerating cells could be seen in the medial dorsal one-third and caudal part of the striatum (caudate putamen). Conclusion: Massive neurodegeneration are demonstrated by FJC staining in the striatum of Pilocarpine-treated mice, suggesting that the striatum may play a role in recurrent seizure mechanism of chronic epilepsy.
