[摘要] 目的 建立复方鬼臼毒素凝胶中鬼臼毒素有光物质的高效液相测定法。 方法 采用高效液相色谱法，色谱柱为Kromasil ODS柱(4.6 mm×250 mm，5 μm)，流动相为磷酸盐溶液(取磷酸氢二钾17.12 g，置1 000 mL量瓶中，加水稀释至刻度，摇匀，用稀磷酸调pH值至7.5)-甲醇(48∶52)；流速为1.0 mL/min；检测波长为292 nm；柱温为30℃。 结果 鬼臼毒素最低检测限为1.22 ng(S/N=3)，与杂质的分离度符合要求，有关物质检查重复性试验符合要求。 结论 本法简便、准确、快速、专属性强，可用于复方鬼臼毒素凝胶中鬼臼毒素有关物质的控制。

[Abstract] Objective To establish the HPLC method for determination related substances of Podophyllotoxin in Compound Podophyllotoxin Gel. Methods Kromasil ODS-C18 (4.6 mm×250 mm, 5 μm) was used as stationary phase. The mobile phase was the solution containing Phosphoric acid salt solution (Dipotassium hydrogen phosphate 17.12 g, dissolved with the 1 000 mL water, shaking to adjust the pH value to 7.5 with the thin Phosphoric acid) - methanol (48∶52); flow rate was 1.0 mL/min, wavelength of UV detection was 292 nm, and temperature of column was 30℃. Results The detection limits was 1.22 ng (S/N=3), which met the requirement with the impurity resolution and the repeatability for related substances for Podophyllotoxin. Conclusion This method is simple, accurate, quick and very good specificity, and can be applied to the determination related substances of Podophyllotoxin in Compound Podophyllotoxin Gel.

高纯度的鬼臼毒素治疗生殖器疣疗效显著，且无系统性毒副作用。作为目前治疗尖锐湿疣的一线药物，鬼臼毒素酊剂在临床已应用多年，其治疗作用得到广泛的肯定。酊剂在使用过程中会对皮肤产生较大的刺激性，应用后局部皮肤黏膜糜烂，给患者造成较大的痛苦。为提高患者的依从性，临床使用时会联合局麻药，比如盐酸利多卡因等，以减少鬼臼毒素的刺激性，从而增加患者的依从性。本公司通过摸索应用现代制剂技术将鬼臼毒素和盐酸利多卡因两个药物进行组合研制出复方鬼臼毒素凝胶剂，该剂型比现有的酊剂、膏剂、激光治疗有明显应用优势，大大提高了患者的依从性。以往文献中均报道鬼臼毒素的各种含量测定方法，但无有关物质方面的研究，实际鬼臼毒素受pH影响，易被氧化，本制剂中除含有凝胶剂的常用辅料，还含有盐酸利多卡因局麻药，为考察本复方中鬼臼毒素是否稳定，并且不受盐酸利多卡因的干扰，笔者通过摸索建立了鬼臼毒素的有关物质测定方法，从而更好地评价含鬼臼毒素的各种制剂的质量。

高效液相色谱仪(岛津：型号LC-20AT，LC-solution工作站，SPD-20A检测器，CBM-20A控制器)，CX-250型超声波清洗器(北京市医疗设备二厂)。
