[摘要] 目的 建立克银颗粒中靛蓝和靛玉红含量的测定方法。 方法 采用HPLC法定量分析，色谱柱为Phenomenex C18(250 mm×4.6 mm，5 μm)，流动相为甲醇-水(80∶20，V/V)，流速为1.0 mL/min，柱温为30℃；靛蓝的检测波长为287 nm，靛玉红为292 nm。 结果 靛蓝在0.329～32.900 μg/mL范围内线性关系良好，Y = 56.94C-1.749(r = 0.999 8)，靛玉红在0.390～39.000 μg/mL(r = 0.999 9)范围内线性关系良好，Y = 119.1C+19.71(r = 0.999 9)，其平均回收率分别为99.89%(RSD = 1.00%，n = 5)、99.18%(RSD = 0.48%，n = 5)。 结论 所建立的方法简便、准确、重复性好，可用于克银颗粒的质量控制。

Determination of indigo and indirubin in Keyin Granule by RP-HPLC

[Abstract] Objective To determine the contents of indigo and indirubin in Keyin Granule. Methods The contents of indigo and indirubin were determined by HPLC. The chromatographic column was Phenomenex C18 (250 mm×4.6 mm, 5 μm), the mobile phase was methanol-water (80∶20, V/V), the flow rate was 1.0 mL/min and the temperature of column was 30℃ with detection wavelength of 287 nm for indigo and 292 nm for indirubin. Results The linear correlation of the indigo was in the range of 0.329-32.900 μg/mL [Y = 56.94C-1.749 (r = 0.999 8)] and the recovery was 99.89% with RSD of 1.00% (n = 5). The linear correlation of the indirubin was in the range of 0.390-39.000 μg/mL [Y = 119.1C+19.71 (r = 0.999 9)] and the recovery was 99.18% with RSD of 0.48% (n = 5). Conclusion The method can be applied to determine the contents of indigo and indirubin in Keyin Granule. It can ensure the stability of the quality control of Keyin Granule.

克银颗粒是由青黛、苦参、白花蛇舌草等九味药组成的复方制剂，该药具有清热解毒、凉血活血、祛风止痒的功效，是第四军医大学唐都医院皮肤科治疗银屑病的经验方。处方中君药青黛具有清热解毒、凉血消斑、泻火定惊的功能，用于温毒发斑、血热吐衄、胸痛咳血、口疮、痄腮、喉痹、小儿惊痫[1-2]。青黛的主要有效成分是靛蓝和靛玉红，为了有效控制该制剂质量，本文采用反相高效液相色谱法同时测定克银颗粒中青黛主要药效成分靛蓝和靛玉红的含量 ......
