芍药内酯苷、芍药苷对慢性束缚应激肝郁模型大鼠海马单胺类神经递质及cAMP、cGMP的影响(1)

     摘要目的:观察肝郁模型大鼠脑内单胺类神经递质、环核苷酸(cAMP、cGMP)水平变化,探讨白芍有效成分芍药内酯苷、芍药苷的作用机制及其“柔肝解郁”功效物质基础。方法:以孤养结合慢性束缚应激的方法建立大鼠肝郁模型,用高效液相-电化学(HPLC-ECD)方法检测其海马单胺类神经递质水平,酶联免疫吸附试验(ELISA)测定cAMP、cGMP水平,观察模型组大鼠给药前后的变化。结果:与正常组比较,模型组大鼠脑内单胺类神经递质肾上腺素(E)、去甲肾上腺素(NE)、多巴胺(NA)、5-羟吲哚乙酸(5-HIAA)水平明显降低(P<0.001或P<0.01),cAMP水平降低、cGMP水平升高(P<0.01或P<0.01);与模型组比较,芍药内酯苷、芍药苷高剂量组能明显增加模型组大鼠海马内单胺类神经递质水平,芍药苷高剂量组可提高模型组大鼠海马cAMP水平(P<0.05),芍药内酯苷高剂量、芍药苷低剂量组可显著降低模型组大鼠海马cGMP水平(P<0.05)。结论:芍药内酯苷、芍药苷作为白芍的主要有效成分,可能通过对脑内单胺类神经递质及环核苷酸的调节作用来抵抗肝郁状态,发挥其“柔肝解郁”作用。

    AbstractObjective:To observe the changes of monoamine neurotransmitters and cAMP, cGMP of liver depression model rats caused by chronic immobilization stress, and to study the mechanism of Albiflorin and Paeoniflorin and material basis of “emolliating liver to resolve depression”.Methods:The model was induced by chronic immobilization stress and solitary.HPLC-ECD was used to analyze the levels of monoamine neurotransmitters.The contents of cAMP and cGMP were detected by ELISA.The changes of the model group were observed before and after administration.Results:Compared with normal group,the levels of monoamine neurotransmitters, noradrenaline, dopamine and 5-hydroxyindoleacetic acid in the model rats decreased significantly (P<0.001 or P<0.01).The content of cAMP decreased, while the content of cGMP increased (P<0.01 or P<0.01); Compared with model group, the high dose of Albiflorin and Paeoniflorin can increase the levels of monoamine neurotransmitters significantly (P<0.05).The high dose of Paeoniflorin can increase the content of cAMP (P<0.05), while the content of cGMP were decreased significantly in high dose of Albiflorin and low dose of Paeoniflorin (P<0.05).Conclusion:Albiflorin and Paeoniflorin as the main ingredient of Radix Paeoniae Alba, may resist the state of liver depression through the regulation of cAMP, cGMP and monoamine neurotransmitters in brain, so as to play the role of emolliating liver to resolve depression.

    白芍(Paeonia lactiflora Pall.),味苦、酸,性微寒,归肝、脾经。甘补酸收,有补血敛阴,柔肝缓急之功,既能养肝血,又能柔肝体,最能顺应肝之特性,为养血柔肝的代表[1]。芍药内酯苷、芍药苷为白芍的2种有效成分[2],研究表明,白芍提取物、芍药苷、芍药内酯苷均具有抗抑郁作用[3-5],其中白芍提取物、芍药苷作用机制多与调节相关信号通路[6]、下丘脑-垂体-肾上腺皮质轴(HPA轴)和增强单胺类神经递质[7]等相关,但关于芍药内酯苷的作用机制则报道较少。本研究通过复制大鼠慢性束缚应激肝郁模型,通过观察海马组织中单胺类神经递质以及cAMP、cGMP水平变化和芍药内酯苷、芍药苷对其干预作用,侧重研究神经系统的变化,以期深入探讨芍药内酯苷、芍药苷作用机制及白芍“柔肝解郁”功效物质基础。, 百拇医药(赵丹萍 张建军 王旭 王小花 王成龙 王玉杰 王林元)
