红藻氨酸致癫痫大鼠腺苷酸激酶2表达变化的研究
王晶+殷亮++++++吕涌涛++++++冯肖亚[摘要] 目的 研究红藻氨酸(KA)致大鼠颞叶癫痫(TLE)后腺苷酸激酶2(AK2)的表达变化,探讨AK2参与颞叶癫痫的发病机制。 方法 将60只雄性SD大鼠分为正常组(10只)和模型组(50只),模型组建立TLE模型;按致痫后6 h、12 h、1 d、3 d、1周对模型组再次进行分组,每组各10只。利用半定量RT-PCR法及Western bloting技术检测AK2 mRNA及蛋白在大鼠额叶及海马中的表达变化。 结果 致痫后额叶及海马组织中AK2 mRNA及蛋白表达均较正常组明显升高(P < 0.05);致痫后3 d组AK2 mRNA及蛋白含量达到高峰,致痫后1周逐渐降低(P < 0.05)。 结论 AK2可能参与癫痫的发生机制,为癫痫发病机制提供新的依据。

[Abstract] Objective To determine the expression levels of multifunction cytokine AK2 in kainic acid-induced epileptic rat, and to investigate whether AK2 participates in the pathogenesis of EP. Methods 60 male SD rats were divided into control group (10 cases) and model group (50 cases), the animal model of temporal lobe epilepsy was established in model group by intra cerebroventricular injection of KA. Then the rats in the model group were divided into 6 hours group, 12 hours group, 1 day group, 3 days group and 1 week group, with 10 cases in each group, according to the different time points after seizures.
