棓丙酯对大鼠肝纤维化防治作用的实验研究(1)

     【摘要】 目的 观察棓丙酯对四氯化碳导致的肝纤维化大鼠的保护作用,为临床治疗肝纤维化提供实验依据。方法 以四氯化碳皮下注射复制大鼠肝纤维化模型,设立正常对照组、肝纤维化模型组和棓丙酯组,棓丙酯组在造模的同时给予棓丙酯注射液皮下注射。6周后取肝组织常规HE染色观察肝脏病变,天狼猩红胶原染色、肝组织羟脯氨酸(HYP)含量测定观察肝纤维化程度;赖氏法测定血浆丙氨酸氨基转移酶(ALT);TBA 法检测肝组织丙二醛(MDA)水平,产色基质偶氮法鲎试剂定量测定血浆内毒素。结果 棓丙酯与肝纤维化模型组比较,①肝脏的损伤性改变较轻,肝组织HYP 及肝纤维化指数(FI)明显降低。②血浆内毒素含量、ALT 及肝组织MDA 均有不同程度的降低。结论 棓丙酯具有一定的抗肝纤维化的作用。

    Preventive effect of Propylgallate on experimental hepatic fibrosis in rats

    【Abstract】 Objective

    To explore the effects of propylgallate on hepatic fibrosis in rats. Methods Hepatic fibrosis in rats was induced by carbon tetrachloride, 24 rats were randomly divided into normal control group, hepatic fibrosis group and propylgallate group, propylgallate group rats were given propylgallate subcutaneously while inducing hepatic fibrosis by carbon tetrachloride. Rats were killed for collecting blood and liver tissues in 6 weeks. The pathological changes were observed by hematoxylineosin staining, the degree of liver fibrosis was observed by stained of Sirius red for collagen and the contents of hydroxyproline in liver tissues. Plasma alanine aminotransferase (ALT) was determined with ReitmanFrankel method; the content of malondialdehyde (MDA) in liver tissue was determined with TBA method. Endotoxin in blood plasma was measured with a limulus amebocyte lysate test kit. Results Propylgallate group compared with the hepatic fibrosis model group: ① fibrotic changes in liver were ameliorated, hydroxyproline in liver tissue and hepatic fibrosis index were significantly reduced. ② Plasma endotoxin, ALT and liver MDA were reduced as well. Conclusion Propylgallate has significant protective effect on liver fibrosis in rats induced by carbon tetrachloride.

    赤芍为毛莨科植物芍药、草芍药及川赤芍的干燥根,味苦性微寒,入肝经。研究显示赤芍具有广泛药理作用,如抗炎、护肝及抗氧化作用[1]。棓丙酯系由中药赤芍的活性成分没食子酸经酯化反应成没食子酸丙酯,为全新化学合成药物,具有更强的生物效应,有抗血小板聚集、抗炎、抗自由基、扩张血管和改变血流流变、改善微循环等作用,临床上多用来预防与治疗脑血栓、冠心病及外科手术后的并发症血栓性深静脉炎等。棓丙酯的这些药理作用与肝纤维化的发病机理关系密切,但目前尚未见其抗肝纤维化作用的研究。本研究通过观察棓丙酯对实验性肝纤维化形成过程及血浆内毒素的影响 ......
