浙江省萧山区轮状病毒A组G型和P型基因同源性比较与进化分析(1)

     [摘要] 目的 分析浙江省萧山区婴幼儿急性腹泻患者中分离的轮状病毒A组G型、P型的基因序列,探讨该病毒的遗传变异和分子特性。方法收集2010年6~12月浙江省萧山区急性婴幼儿(≤5岁)腹泻患者粪便标本,应用多重PCR技术从临床标本中扩增轮状病毒A组G型、P型基因片段并测序分析,比较基因的亲缘关系并绘制进化树。结果 162例粪便样本,ELISA结果为54例阳性。PCR检测结果 G1型为32株,G2为5株,G3为9株,P型中,P4为3株,P8为5株,成功地对16例A组轮状病毒基因进行测序。2010年A组轮状病毒基因的核苷酸序列与参考株具有较高的同源性,且A组基因核苷酸序列发生了一定的变异。结论 轮状病毒A 是引起浙江萧山地区急性腹泻的主要的病原之一,其病原具有基因型别的多样性,G1型可能是2010年浙江萧山地区流行的优势株。

    Analysis of Homology and Evolution of G and P Genotype of Rotavirus A Group in Xiaoshan of Zhejiang Province

    [Abstract] Objective To analyze the gene sequence of rotavirus A group of G and P genotype from acute diarrhea patients in Xiaoshan district of Zhejiang Province and explore the virus genetic variation and molecular characteristics. MethodsCollect stool specimens of infants(≤5 years)with acute diarrhea in Xiaoshan district of Zhejiang Province from Jun 2010 to Dec 2010, and G and P genes of rotavirus A group from clinical specimens were amplified by multiplex RT-PCR, the amplified products with expected size were submitted to direct sequencing. Genetic factors were analyzed and drawn phylogenetic trees. Results Fifty-four cases of rotavirus A group gene were amplified from 162 cases of stool samples. 47 cases were G type, and 7 cases were P type. 32 strains were G1 type, 5 strains were G2 type, 9 strains were G3 type; 3 strains were P4 type, 5 strains were P8 type. 16 products with positive samples were successfully sequenced. After alignment, high homologies of amino acids of G and P genes were observed among sequences of rotavirus A group virus. However, some mutations of rotavirus A group were found in 2010. ConclusionRotavirus A viruses were one of the major pathogens causing acute diarrhea of infants (≤5 years), multiple genotypes of Rotavirus A were found in the specimens, and G1 genotype was probably identified as the prevalent strain in Xiaoshan, Zhejiang province in 2010 ......
