XAF1蛋白在人肝癌组织中的表达及术后复发中的作用研究(1)

     [摘要] 目的 XIAP相关因子1(XIAP-Associated Factor 1,XAF1)可与XIAP直接结合并拮抗其抗凋亡作用,本文研究了XAF1在人肝细胞癌组织中的表达,并探讨了其在肝癌患者预测复发中的价值。 方法 采用免疫组化法检测38例HCC患者肝癌组织中XAF1蛋白的表达,并分析其与术后复发的关系。 结果 XAF1在人肝癌组织中表达免疫组化实验结果提示,XAF1在人肝癌组织中的表达阳性率为36.84%(14/38),在癌旁正常组织中的表达阳性率为100%(38/38),两者差异有统计学意义(P<0.05)。24例XAF1低表达患者中,XAF1表达水平与PVTT(P=0.012)、AFP高水平(P=0.016),肿瘤大小(P=0.037)有关;Log-rank检验结果显示,XAF1低表达患者的术后1年无瘤生存率显著低于XAF1正常表达患者(P<0.05),是影响HCC术后1年生存率的因素之一。 结论 XAF1在人体肝癌组织中表达水平低于癌旁正常组织,这可能是影响肝癌术后复发的一个潜在危险因素。

    Role of XAF1 in tumor recurrence of patients with hepatocellular carcinoma treated with hepatectomy

    [Abstract] Objective XIAP-associated factor 1 (XAF1) is a putative tumor suppressor that exerts its proapoptotic effects through both caspase-dependent and–independent means. In this study we investigated the expression XIAP-associated factor 1 (XAF1) in human hepatocellular carcinoma (HCC) and its mechanism of post-surgical recurrence of patients with HCC. Methods Immunohistochemistry was used to detect the expression of XAF1 in 38 patients with HCC after hepatectomy, and the relationship between the expression of XAF1 and the post-surgical recurrence of patients with HCC was analyzed by bioinformatics. Results Immunohistochemisitry results showed that the positive ratio was 36.84% (14/38) and 100% (38/38) in human HCC tumor tissues and corresponding normal liver tissues significantly, and there was significant difference between them (P<0.05). HCC patients with low expression of XAF1 were associated with PVTT (P=0.012), highalpha-fetoprotein (AFP) level (P=0.016) and tumor size (P=0.037) , respectively. It was demonstrated that the 1-year tumor-free survival rate of normal XAF1 expression patients was significantly higher than that of low XAF1 expression patients (P<0.05), suggesting that low XAF1 expression may be associated with the postoperative recurrence of HCC patients.
