大鼠脑血肿周围Caspase-3的表达与神经细胞凋亡的关系(1)

     [摘要] 目的 研究大鼠脑血肿周围脑组织Caspase-3的表达和神经细胞凋亡及探讨二者之间的关系。 方法 60只Wistar大鼠随机分为假手术组(30只)和出血组(30只)。采用大鼠自体股动脉血注入尾壳核建立脑出血模型,分别于术后6、12、24、48、96 h 5个时间点取脑,应用免疫组化方法检测血肿周围组织Caspase-3的表达,TUNEL法检测血肿周围神经细胞凋亡。 结果 出血组大鼠脑血肿周围组织Caspase-3的表达及细胞凋亡明显高于假手术组(P < 0.01),Caspase-3的表达与血肿周围神经细胞凋亡呈正相关性(r = 0.956,P < 0.05)。 结论 Caspase-3、神经细胞凋亡参与了大鼠脑血肿周围脑组织损伤的病理过程。

    The association between expression of Caspase-3 and neuron apoptosis in perifocal tissue of intracerebral hemorrhage in rats

    [Abstract] Objective To research the expression of Caspase-3 in perifocal tissue of intracerebral hemorrhage (ICH) and investigate the association between Caspase-3 and neuron apoptosis in perifocal tissue of ICH. Methods Sixty wistar rats were randomly divided into ICH group (n = 30) and sham operated group (n = 30). ICH rat models were prepared with injection of autologous blood into the right caudate nucleus. Each group was randomly divided into 5 various stages: 6 h, 12 h, 24 h, 48 h and 96 h. The expression of Caspase-3 was measured with immunohistochemistry method. Apoptosis was characterized by terminal deoxynucleotide transferase mediated uridine 5-triphosphate-biotin nick end-labeling (TUNEL) staining. Results Protein expression of Caspase-3 and neuron apoptosis of ICH group was higher than sham operated group (P < 0.01). The association between Caspase-3 and neuron apoptosis in perifocal tissue of ICH was positively relative (P < 0.05). Conclusion Caspase-3 and apoptosis are involved in pathophysiologic process of ICH.
