依达拉奉对大鼠脑缺血再灌注损伤的保护作用研究(1)

     [摘要] 目的 探讨依达拉奉对大鼠脑缺血再灌注损伤的保护作用。方法 将36只SD大鼠随机分为假手术组、NS组及依达拉奉组,制备SD脑缺血再灌注模型,依达拉奉组在造模后即刻和12h分别给予依达拉奉腹腔注射,假手术组和NS组分别给予生理盐水腹腔注射,比较3组神经功能缺损评分及SOD、MDA含量。结果 与假手术组比较,NS组与依达拉奉组神经功能缺损评分分别为(2.74±0.56)、(1.38±0.22)分均明显升高(P<0.05),而与NS组比较,依达拉奉组神经功能缺损评分则明显下降(P<0.05);与假手术组比较,NS组与依达拉奉组SOD活性分别为(113.29±7.61)、(144.93±10.48)U/mL明显下降(P<0.05),而与NS组相比较,依达拉奉组SOD活性明显增高(P<0.05);与假手术组比较,NS组与依达拉奉组MDA水平分别为(3.71±1.24)、(2.25±0.98)nmol/mL明显升高(P<0.05),而与NS组相比较,依达拉奉组MDA水平明显下降。结论 依达拉奉对大鼠脑缺血再灌注损伤具有明显的保护作用。

    [Abstract] Objective To investigate the protective effect of edaravone on cerebral ischemia reperfusion injury in rats. Methods 36 SD rats were randomly divided into sham operation group, NS group and edaravone group, preparation of SD in cerebral ischemia reperfusion model group, edaravone immediately after modeling and 12 h were given intraperitoneal injection of edaravone, sham operation group and NS group were given intraperitoneal injection of normal saline, neural function defect score and SOD, the content of MDA of 3 groups. Results The Compared with the sham operation group, NS group and edaravone group, the score of neural function defect were (2.74±0.56), (1.38±0.22) were significantly increased (P<0.05), compared with NS group, the score of neural function defect edaravone group decreased significantly (P<0.05); compared with the sham operation group NS group and edaravone group, the activity of SOD were (113.29±7.61), (144.93±10.48) U/ml decreased significantly (P<0.05), and compared with the NS group, edaravone group SOD activity obviously increased (P<0.05); compared with the sham operation group, NS group and edaravone group MDA horizontal don't as (3.71±1.24), (2.25±0.98) nmol/ml increased significantly (P<0.05), and compared with the NS group, the levels of MDA of edaravone group decreased significantly. Conclusion edaravone on cerebral ischemia reperfusion injury has obvious protective effect.

    近年来的研究表明自由基的连锁反应是脑缺血的核心病理环节,过多的自由基在脑缺血再灌注损伤中占据极其重要地位,可加重脑缺血损伤而导致神经功能损害[1]。依达拉奉是一种由日本Mitsubishi-Tokyo公司研制开发的新型自由基清除剂,其血脑屏障通透性高,可通过清除自由基、抑制脂质过氧化反应等来发挥强大的脑缺血保护作用,目前认为依达拉奉的作用机制极为复杂,很多机制仍未完全阐明[2]。该研究选取2012年6月—2013年2月间健康雄性SD大鼠为研究对象,主要从抗自由基损伤方面来初步探讨依达拉奉对脑缺血再灌注损伤的保护作用,为该药物在脑缺血治疗上提供参考,现报道如下。

    1 资料与方法

    1.1 一般资料

    选取健康雄性SD大鼠共36只,体重250~300 g。将大鼠随机分为3组,每组12只,分别为假手术组、NS组及依达拉奉组。依达拉奉注射液由江苏先声药业有限公司提供,超氧化物歧化酶(SOD)、丙二醛(MDA)试剂盒均由上海杰美基因医药科技有限公司提供。, 百拇医药(韩春辉)
