雷公藤内酯醇对EAE小鼠脊髓TNF-α\IL-12 和TGF-β1表达的影响(1)

     [摘要] 目的:观察免疫抑制剂雷公藤内酯醇(Tri)对实验性自身免疫性脑脊髓炎(EAE)小鼠模型脊髓组织TNF-α、IL-12 和TGF-β1的影响。方法:采用MOG33-35、佐剂CFA、百日咳毒素免疫动物建立EAE模型,腹腔注射Tri治疗,制作脊髓石蜡切片行HE染色及LBF髓鞘染色,免疫组化染色观察TGF-β1、TNF-α和IL-12的表达,并行相关图像分析。结果:HE染色可见EAE组脊髓有“袖套样”炎性细胞浸润。LBF髓鞘染色可见EAE组小鼠脊髓髓鞘脱失。与佐剂组相比,EAE组TNF-α、IL-12表达增高(P<0.01或P<0.05), TGF-β1表达降低(P<0.05)。与EAE组相比,Tri治疗组TNF-α、IL-12表达降低(P<0.05),TGF-β1表达增高(P<0.05)。结论:Tri可明显改善EAE小鼠脊髓组织病理状况,可能与抑制TNF-α和IL-12等促炎性细胞因子,促进TGF-β1等保护性细胞因子的分泌有关。

    Effects of Triptolide on TNF-α、IL-12 and TGF-β1 in spinal cord ofEAEmice

    [Abstract] Objective: To observe the effects of Triptolide on TGF-β1、TNF-α and IL-12 in spinal cord ofEAEmice. Methods: EAE model was established with MOG35-55, Tri was intraperitoneally injected in treatment group, the spinal cord of mice were disseted and removed for paraffin section. These sections were used for HE, myelin staining and immunohistochemical stain on TGF-β1、TNF-α and IL-12. Results: For EAE models, HE stain showed typical “muff-like” inflammatory cell infiltration, and myelin staining showed demyelination in spinal cord. Compared with adjuvant group, the level of TNF-α and IL-12 in EAE group was increased (P<0.01or P<0.05), and the level of TGF-β1 was decreased(P<0.05). Compared with EAE group, TNF-α and IL-12 in Tri group was decreased (P<0.05), and the level of TGF-β1 was increased significantly(P<0.05). Conclusion: Tri can treat EAE mice effectively. The mechanism may be caused by depression of secretion of TNF-α, IL-12 and promotion ofTGF-β1.
