川芎嗪对人肝星状细胞TGF-β1信号传导的影响(1)

     [摘要] 目的:探讨中药单体川芎嗪(TMP)对肝星状细胞(HSC)转化生长因子-β1(TGF-β1)信号传导的影响。通过检测TGF-β1及其Ⅰ型和Ⅱ型受体、Smad 3及Smad 7 mRNA表达的变化,探讨川芎嗪抗肝纤维化作用的相关机制。方法:体外培养人肝星状细胞, RT-PCR法检测HSC中TGF-β1及其Ⅰ型和Ⅱ型受体、Smad 3及Smad 7 mRNA表达。结果:与阴性对照组相比,TMP A、B、C各组均可显著降低TGF-β1 mRNA(P<0.01);TMP B、C组TβRⅠmRNA相对表达量降低,差异有统计学意义(P<0.05或P<0.01);TMP B、C浓度组的TβRⅡmRNA、Smad 3 mRNA表达量显著降低(P<0.01);TMP B、C组可显著升高Smad 7 mRNA表达量(P<0.01)。结论:川芎嗪可降低HSC中TGF-β1及其Ⅰ、Ⅱ型受体及Smad 3 mRNA表达,同时促进Smad7 mRNA表达,表明川芎嗪可抑制肝星状细胞的活化、增殖,这可能与阻断TGF-β/Smad通路的信号传导有关。

    [Abstract] Objective: To investigate the effects of tetramethylpyrazine on Signal Transduction Induced by Transforming growth factor-β1 in Human Hepatic Stellate Cells. To explore the anti-hepatic fibrosis mechanisms of tetramethylpyrazine by studying the expressions of TGF-β1 mRNA, typeⅠ and typeⅡ TGF-β receptor mRNA, Smad 3 and Smad 7 mRNA. Methods: HSCs cultured in vitro were divided into negative control group, positive control group(cultured with Matrine 500 mg/L), experimental group(cultured with different concentrations of TMP (50 mg/L、100 mg/L、200 mg/L、400 mg/L、800 mg/L). The effect of TMP was evaluated of the level of TGF-β1, TβRⅠ, TβRⅡ, Smad 3 and Smad 7 mRNA by RT-PCR.
