白花蛇舌草总黄酮的超声提取工艺研究(1)

     [摘要] 目的:优选白花蛇舌草总黄酮超声提取最佳工艺。方法:选择无毒的有机溶剂乙醇作为提取溶剂,超声波作为提取方法,然后用聚酰胺色谱柱纯化总黄酮成分,采用NaNO2-Al(NO3)3-NaOH显色法,以芦丁为对照品计算总黄酮的含量,并用正交设计法进行提取工艺优化。通过考察不同因素和水平对提取效率的影响,优选出白花蛇舌草中总黄酮的最佳提取工艺。结果:最佳提取工艺是先用石油醚脱脂1次,提取溶剂为70%乙醇,料液比为1:20,超声时间为30 min,提取次数为3次,提取率为2.375 6%。结论:优选出的工艺简单易行,能保证白花蛇舌草中总黄酮提取完全。

    Study on the ultrasonic extraction technique of the total favonoids from hedyotis diffusa wild

    [Abstract] Objective: To establish the best ultrasonic extraction technique of the total favonoids from hedyotis diffusa. Methods: Chose non-toxic organic solvents ethanol as solvent extraction, used ultrasonic extraction as method, and then used as polyamide chromatography column purification flavonoid, using NaNO2-Al(NO3)3-NaOH show color method, rutin ethanol-water calculation, the content of flavonoids with orthogonal design method for the extraction process optimization. By investigating the different factors and level on the influence of the optimum extraction efficiency, the white snake tongue grass of the best extraction technology of flavonoids. Results: The best condition was as follows: degreasing with petroleum ether fot two times, then add 20 times volume of 70% alcohol, ultrasonic extract for 30 min with 3 times. The extration yield was 2.375 6%. Conclusion: The extraction method is appropriate, simple and feasible. It is an ideal way to extract the flavonoids of hedyotis diffusa.

    白花蛇舌草(hedyotis diffusa)为茜草科一年生草本植物白花蛇舌草的全草,具有清热解毒、利尿消肿之功效,是民间常用的中草药[1]。现代药理研究表明,其含有的总黄酮具有抗菌、抗炎、抗肿瘤、增强免疫力等作用,是一类极具开发价值的天然活性物质[2~8]。正交设计法(orthogonal design)是一种研究与处理多因素实验的重要数理统计方法,可以尽可能少的试验次数,获得最满意的试验结果。因此,本实验采用正交设计法,通过考察不同因素、不同水平对总黄酮提取率的影响 ......
