黑曲霉Aspergillus niger对人参皂苷Re的微生物转化(1)

     [摘要] 目的:筛选长白山人参土壤中的活性微生物,转化单体人参皂苷产生稀有人参皂苷成份。方法:从长白山人参根际土壤中分离各类菌株,对单体人参皂苷Re进行微生物转化,通过硅胶柱层析等方法对转化产物进行分离纯化,采用波谱解析及理化常数对其进行结构鉴定。结果:从长白山人参根际土壤中分离各类真菌菌株68株,3株真菌对三醇组人参皂苷Re具有转化作用,其中黑曲霉Aspergillus niger的转化活性较强,转化产物为人参皂苷Rg1、Rg2和Rh1。结论:首次报道黑曲霉能将人参皂苷Re转化为人参皂苷Rg1、Rg2和Rh1这一转化过程。

    Microbial transformation of Ginsenoside Re by Aspergillus niger

    [Abstract] Objective: Samples of Ginseng root soils, collect from Changbai Mountain, are used to screen active microorganism which can transform Ginsenoside Re, so as to obtain rare anti-tumor components. Methods: The strains were isolated and screened on liquid transfer medium and yield transfer process for active strains. Then the active strains were tested for their biotransformation properties by using Ginsenoside Re. The biotransformation products were separated and purified through different column chromatographies and identified by spectral analysis and physical constants. Results: Total 68 fungal strains were isolated and 3 active strains showed positive activity on Ginsenoside Re. One strain, Aspergillus niger, was found to have the strong activity on Re. Conclusion: This is the first report on the transformation G-Re to G-Rg1, G-Rg2, and G-Rh1 by microorganism Aspergillus niger.

    人参及其制品中的主要活性成分是人参皂苷,由于人参皂苷分子结构中糖基侧链的不同而显示出不同的性质和药理活性[1-2]。例如人参皂苷Rb1有促进神经细胞生长的作用、降低细胞内钙、抗氧化、清除自由基和改善心肌缺血再灌注损伤等作用[3];人参皂苷Rd能促进T细胞增殖,提高天然杀伤细胞(NK)的活性[4];20(S)-原人参二醇苷元(PPD)具有抗癌活性[5]等。人参皂苷Re是抗心律失常有效成分,可抑制吗啡诱发小鼠产生的耐药性等作用;人参皂苷Rg1具有使中枢神经兴奋、抗疲劳、改善记忆、学习功能等作用[6];人参皂苷Rg2可抑制兔血小板释放反应等作用 ......
