不同提取工艺下甘草提取物中甘草酸铵含量测定研究(1)

     [摘要] 目的:建立甘草提取物中甘草酸铵的含量测定方法,优选甘草酸铵的提取工艺。方法:以甘草酸铵为指标,采用高效液相色谱法测定甘草酸铵含量;比较煎煮法、水提醇沉法、0.5%氨水提取与70%乙醇超声法等工艺的差异。结果:0.5%氨水提取甘草酸铵含量较高,甘草酸铵含量在5.267 5~337.12 μg范围里呈较好的线性关系;甘草酸铵的回收率为98.51%。结论:0.5%氨水提取工艺所得甘草提取物中甘草酸铵含量较高,甘草酸铵转移率最高,提取方法合理、可行、效率高。

    Study on content determination of Monoammonium Glycyrrhizinate in the distilled licorice by different extraction technique

    [Abstract] Objective: To establish a method for content determination of Monoammonium Glycyrrhizinate in the distilled licorice and to optimize the extraction technique. Methods: Taking Monoammonium Glycyrrhizinate as index, we used the technique of HPLC to determine its content. Then compared the difference among water-decocting water-extracting and alcohol-depositing; 0.5% ammonia-extracting and 70% alcohol ultrasonic-method. Results: The content from 0.5% ammonia-extracting extraction of Monoammonium Glycyrrhizinate was the higher. In the range of 5.267 5-337.12 μg, the Monoammonium Glycyrrhizinate showed good linear relationship. The recovery of Monoammonium Glycyrrhizinate was 98.51%. Conclusion: The content from 0.5% ammonia-extracting extraction of Monoammonium Glycyrrhizinate is the higher. The transfer rate of Monoammonium Glycyrrhizinate is the highest. The 0.5% ammonia-extracting is reasonable, feasible and efficient.

    甘草为豆科植物甘草(Glycyrrhiza uralensis Fisch)、胀果甘草(Glycyrrhiza inflata Bat)或光果甘草(Glycyrrhiza glabra L)的干燥根及根茎。具有补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药的作用。主要用于脾胃虚弱,倦怠乏力,心悸气短,咳嗽痰多,脘腹、四肢挛急疼痛,痈肿疮毒,缓解药物毒性、烈性[1]。在中成药生产中,常以甘草的提取物入药。其主要活性成分为甘草酸铵(glycyrrhizic acid,GA),对许多疾病有预防和治疗作用。甘草酸铵也是甘草甜味的有效成分,它的特点是高甜度、低热能、安全无毒,起泡性和溶血性作用低。能与多种生物碱、抗生素、氨基酸等生成复盐或复方制剂,具有协同、增溶、增加药物稳定性、提高生物利用率及降低毒副作用的功效。同时,还广泛用于食品添加剂、化妆品、烟草行业[2]。我公司小儿七星茶颗粒成方制法中的甘草也是以提取物入药,它的提取工艺与纯化方法直接影响该产品的质量及溶化性。因此,研究甘草提取物的提取工艺及纯化方法具有重要意义。本实验通过优选甘草的提取工艺 ......
