斑贞1号、川芎嗪和氟尿嘧啶联合对人胃癌细胞多药耐药基因1表达的影响(1)

     [摘要] 目的 探讨斑贞1号、川芎嗪(TMP)和氟尿嘧啶(5-FU)联合逆转人胃癌细胞系SGC-7901/ADR与多药耐药基因1(MDR1)表达的相关性。 方法 以SGC-7901/ADR细胞为靶细胞,以不加药组为阴性对照组,氟尿嘧啶组、斑贞1号组、斑贞1号+氟尿嘧啶组、斑贞1号+氟尿嘧啶+川芎嗪(TMP)组为实验组,采用半定量RT-PCR法检测SGC-7901/ADR细胞在不同药物作用下MDR1 mRNA的表达情况。 结果 氟尿嘧啶组与阴性对照组相比,MDR1 mRNA表达差异无统计学意义(P > 0.05),而其他药物组与对照组及组间比较差异有统计学意义(P < 0.05),斑贞1号+氟尿嘧啶 + TMP组表达量最少。 结论 斑贞1号、TMP和氟尿嘧啶联合逆转SGC-7901/ADR细胞耐药性可能与MDR1基因相关,为当前胃癌的治疗提供了新的理论依据。

    Effect of Ban Zhen 1, tetramethylpyrazine and fluorouracil on human gastric cancer cells multidrug resistance gene expression

    [Abstract] Objective To explore the correlativity of Ban Zhen 1, Tetramethylpyrazine (TMP) and fluorouracil reversing human gastric cancer cell line SGC-7901/ADR with Multidrug resistance gene 1 (MDR1) expressions. Methods SGC-7901/ADR cells was considered as target cells, no medicines group was considered as negative control, Fluorouracil, Banzhen-1, Banzhen-1 + Fluorouracil, Banzhen-1 + Fluorouracil + TMP were considered as experiment groups. The expression of SGC-7901/ADR cells' MDR1 mRNA under different medicine groups was detected by the semi-quantitative RT- PC. Results The expression of MDR1 mRNA of Fluorouracil was the same as negative control (P > 0.05), however, the expression of MDR1 mRNA in other groups were different from each other and the group of Banzhen-1 + Fluorouracil + TMP was lowest (P﹤0.05). Conclusion It is possible correlated to MDR1 for Ban Zhen 1, TMP and Fluorouracil reversed multidrug resistance in human gastric cancer cells, which may supply a new theoretical basis for the clinical effective treatment of gastric carcinoma.

    化疗是治疗胃癌的有效手段之一,但胃癌多药耐药性(MDR)是导致化疗失败的主要原因。近几年发现MDR1在胃癌多药耐药性方面起着重要作用,侯培珍等[1]已证实斑贞1号 + 川芎嗪(TMP) + 氟尿嘧啶对人胃癌SGC-7901/ADR细胞有较强的逆转作用。因此本文笔者在此实验的基础上,通过检测不同药物组MDR1 mRNA的表达量从分子生物的角的进一步探讨斑贞1号 + TMP + 氟尿嘧啶对人胃癌SGC-7901/ADR细胞的作用机制 ......
