桑仙降压颗粒对自发性高血压大鼠CD34和心肌血管抑素蛋白密度表达的干预(1)

     [摘要] 目的 通过桑仙降压颗粒对自发性高血压大鼠(spontaneously hypertensive rats,SHR) CD34和AS蛋白光密度值的干预,探讨其对微血管数目及血管新生抑制因子的作用及机制。 方法 SHR随机分为3组,蒸馏水空白对照组、吲哒帕胺西药对照组、桑仙降压颗粒组,以Wister 大鼠为正常对照,4周及8周后观察SHR心肌AS蛋白光密度及CD34的表达。 结果 相较空白对照组,4周及8周后桑仙降压颗粒组CD34表达均明显增多且心肌AS蛋白光密度值明显降低。 结论 桑仙降压颗粒能有效减轻SHR大鼠微血管损害,其机制之一可能与降低AS蛋白密度表达有关。

    The Intervention study of Sang Xian Jiang Ya Granule on epression of myocardial CD34 and endostatin(AS) protein optical density of spontaneously hypertensive rats

    [Abstract] Objective To observe the effect of Sang Xian Jiang Ya Granule(SXJYG) on spontaneously hypertensive rats(SHR) myocardial CD34 and endostatin (AS) protein optical density values, and investigate the intervention of SXJYG on microvascular number and angiogenesis inhibiting factor. Methods SHR were randomly divided into 3 groups: those gavaged by distilled water as the control group, those gavaged by indapamide as the control group of Western Medicine, SXJYG group, compared with wistar as normal control group. After 4 and 8 weeks, the expression of myocardial AS protein and CD34 were observed. Results Compared with the blank control group, after 4 and 8 weeks SXJG group CD34 expression were significantly increased but myocardial AS optical density value were decreased. Conclusion SXJXG can reduce the SHR rat microvascular damage, and one of its mechanisms may be related to the decrease of AS protein expression in relation to density.

    高血压是最常见的心血管病,是全球范围面临的重大公共卫生挑战。高血压不仅发病率高,还常常引起严重的心脑肾并发症,是心脑血管事件的主要危险因素之一,成为致死、致残的重要原因。可见,大力开展高血压的发病机制、防治研究已刻不容缓[1],研究方法众多,然其机制鲜有从微血管入手,因此本研究通过观察桑仙降压颗粒对SHR心肌CD34及AS的影响,观测其对SHR微血管密度及AS蛋白光密度的干预并作相关机制分析,以期对积极探索预防靶器官损害起到推动作用。

    1 材料与方法

    1.1 材料

    1.1.1 动物与分组 4周龄Wistar大鼠18只,雌雄各半,体重210~250 g,由山东医科大学动物室提供[动物合格证号:SCXK(鲁)20090001]。以A标示正常对照组即Wistar大鼠组;4周龄SHR,雌雄各半,体重200~240 g,由中国医科院阜外心血管医院提供(医动字第01-108号),42只,B、C、D分别标示SHR不同的给药方式,B:SHR给予蒸馏水为空白对照组,C:SHR给予桑仙降压颗粒为实验组,D:SHR给予吲哒帕胺为西药对照组;观测事项点:0组的观测时间为Wistar大鼠、SHR4周龄;1、2的观测时间为Wistar大鼠、SHR给药后4周、8周 ......
