生发酊对C57BL/6小鼠毛囊细胞凋亡的影响(1)

     [摘要] 目的 从毛囊细胞凋亡的角度出发,观察生发酊对动物模型小鼠毛发生长周期的影响,评价其治疗脱发的效果。 方法 将C57BL/6小鼠随机分为生发酊组、阳性对照组、阴性对照组、模型对照组,制备小鼠脱发动物模型。脱毛后第2天,于脱毛区涂相应药物;脱毛后第17天,颈椎脱臼处死小鼠,背部平行脊柱相同部位取材,用原位杂交技术原位末端标记法(TUNEL)检测毛囊内细胞的凋亡状况。 结果 生发酊组[(15.82±0.36)个/视野]与阳性对照组[(16.12±0.39)个/视野]细胞凋亡数量明显少于阴性对照组[(40.32±0.47)个/视野]与模型对照组[(42.16±0.54)个/视野],差异有统计学意义(P<0.01);生发酊组与阳性对照组比较差异无统计学意义(P>0.05)。 结论 生发酊对毛囊细胞凋亡有明显的抑制作用,此为其治疗脱发的可能机制之一。

    Impact of hair-growth tincture on hair follicle cell apoptosis in animal model for C57BL/6 mice

    [Abstract] Objective To observe the impact of hair-growth tincture on mice hair growth cycle in animal model from the angle of hair follicle cell apoptosis and to evaluate its effects in treatment hair loss. Methods C57BL/6 mice were randomly divided into hair-growth tincture group,positive control group,negative control group and model control group and then mice hair loss model were made.Corresponding drugs were smeared to hair loss area from the second day after hair loss.Mice were killed by cervical dislocation at the 17 day after hair loss.The samples were draw from same part of back parallel spine.In situ hybridization TUNEL was used to test the apoptosis condition of hair follicle cell. Results The apoptosis number in hair-growth tincture group and positive control group with a view of (15.82±0.36) cells and (16.12±0.39) cells were obvious less than those in negative control group and model control group with a view of (40.32±0.47) cells and (42.16±0.54) cells,and the differences were statistical significant (P<0.01).The difference between hair-growth tincture group and positive control group had no statistical significance (P>0.05). Conclusion Hair-growth tincture has obvious role in inhibiting hair follicle cell apoptosis and this may be one of the mechanism in treating hair loss.

    脱发是一种常见的疾病,在人群中的发病率较高,其发病机制尚不完全清楚。部分脱发具有先天遗传性,后天可能与精神、内分泌、感染、自身免疫、中毒等因素有关。毛囊是哺乳动物的特征性结构,近年来毛囊生物学受到广大皮肤病学者的广泛关注,毛囊成为研究细胞凋亡极好的生物系统[1],由于其是毛发脱落最关键的器官,研究毛囊的生长特征、改善毛囊的生长环境是治疗脱发疾病的关键。我院皮肤科经过多年的临床研究,研制出用于治疗脱发疾病的自制中药外用制剂——生发酊,该药物经多年临床应用,显示出明显的乌发、生发的作用,效果显著,具有一定的应用价值。本实验目的在于阐明该药治疗多种脱发的作用机制,为临床应用提供一定的实验依据。

    1 材料与方法

    1.1 实验材料

    C57BL/6小鼠,合格证号:SCXK(吉)2014-001,雌雄各半,清洁级,6周龄,体重19~21 g,共80只,由吉林大学实验动物中心提供。常规饲养,待小鼠适应环境后开始实验。, 百拇医药(张春洪)
