EtNBSe联合光动力疗法对成纤维细胞增殖及TGF-β1分泌的影响(1)

     【摘要】目的:观察EtNBSe(5-ethylamino-9-diethylamino benzo[a]phenoselenazinium chloride)联合光动力疗法(photodynamic therapy,PDT)对人包皮成纤维细胞体外增殖及分泌表型的影响。方法:成纤维细胞接种至培养板,采用不同浓度的EtNBSe处理细胞2 h后,给予同一时间剂量的红光辐照,MTT法检测细胞的功能;酶联免疫吸附试验(ELISA)法检测转化生长因子-β1(transforming growth factor-β1,TGF-β1) 水平的变化。结果:(1)与空白组比较,细胞接受EtNBSe-PDT处理后,EtNBSe(60、120 pM)组有增殖活性(P<0.05);(2)与空白组比较,EtNBSe(60、120 pM)组,TGF-β1浓度分别增加28.57%和45.11%(P<0.05)。结论:EtNBSe-PDT可促进体外生长的成纤维细胞增殖活性,诱导成纤维细胞TGF-β1的分泌。

    【Abstract】Objective:To investigate the influences of 5-ethylamino-9-diethylamino benzo[a]phenoselenazinium chloride (EtNBSe) together with Photodynamic Therapy on human foreskin fibroblast cells proliferation in vitro and secretion phenotype.Methods:Inoculate fibroblasts to the culture plates, treated with different concentrations of EtNBSe, and given the same time red light exposure 2 hours after treatment. Using MTT assay examine the cell functions, and ELISA assay examine the changes of TGF-β1.Results:Compared with the control group, EtNBSe-PDT treated cells especially EtNBSe(60 or 120pM) group proliferated obviously(P<0.05); and the concentration of TGF-β1 in these two groups was increased 28.57% and 45.11% respectively(P<0.05).Conclusion:In vitro, EtNBSe-PDT treatments promote the activity of growing fibroblast proliferation, induce TGF-β1 secretion.
