丙泊酚对大鼠单肺通气时代谢酶的影响(1)

     【摘要】 目的:观察丙泊酚对大鼠单肺通气时肺代谢酶的影响,为丙泊酚在胸科麻醉中的应用提供实验研究的数据。方法:40只健康清洁级SD大鼠随机分成四组:P 1组:空白组(灌注液中无丙泊酚);P2 组:含36 mg/L丙泊酚;P3 组:含24 mg/L丙泊酚;P 4组:含12 mg/L丙泊酚,每组10只。按随机顺序取大鼠进行实验,称量体重,麻醉,气管切开插管接呼吸机行单肺机械通气1 h。然后取出肺组织。-70 ℃保存,以备统一作Ca2+ -ATPase、Na+ -k+ ATPase测定。结果:肺组织Na+-K+ATPase活力:P1组通气侧活力显著低于P2、P3、P4 组通气侧(P<0.01);P1 组非通气侧活力显著低于P2、P3 、P 4组非通气侧(P<0.01)。各组非通气侧均低于同组通气侧(P<0.01或0.05)。肺组织Ca2+-ATPase活力:P1 组通气侧显著低于P2、P3、P4组通气侧(P<0.01);P1组非通气侧显著低于P2、P3、P4组非通气侧(P<0.01)。各组非通气侧均显著低于同组通气侧(P<0.01)。结论:通过对大鼠单肺通气时的灌注实验研究发现,丙泊酚对单肺通气时的肺代谢酶有保护作用,其程度与丙泊酚浓度在一定范围内无关。

    【Abstract】 Objective:To investigate the effect of propofol on metabolic enzyme of rats during one-lung ventilation.Methods:40 rats randomly divided into 4 groups(n=10,each):group P1 (no propofol in the perfusate ),group P2 (the concentration of propofol in perfusate was 36mg/L),group P3(24 mg/L),group P4 (12 mg/L). Metabolic enzyme was quantified after one-lung ventilation .After OLV finished ,lung tissue sample were harvest for the criterions of the Na+-K+ATPase activity,Ca2+ATPase activity.Results:(1)The Na+-K+ATPase activity: as compared to the values in group P2、group P3 and group P4 ventilated lung, the Na+-K+ATPase activity was low in group P1 ventilated lung(P<0.01); as compared to the values in group P2、group P3 and group P4airproof lung, the Na+-K+ATPase activity was low in group P1 airproof lung(P<0.01);as compared to the values in reciprocal group ventilated lung, the Na+-K+ATPase activity was low in each group airproof lung(P<0.01 or 0.05).(2)The Ca2+-ATPase activity: as compared to the values in group P2、group P3 and group P4 ventilated lung, the Ca2+-ATPase activity was low in group P1 ventilated lung(P<0.01); as compared to the values in group P2 、group P3 and group P4 airproof lung ......
