金柚提取物对糖尿病肾病大鼠肾组织中p38MAPK炎症信号通路的影响(1)

     【摘要】 目的:研究金柚提取物對糖尿病肾病大鼠肾组织中磷酸化丝裂原活化蛋白激酶p38(p-p38MAPK)及细胞核中核转录因子-κB P65(NF-κBP65)表达的影响。方法:采用一次性腹腔注射链脲佐菌素建立糖尿病大鼠模型,分为正常对照组、糖尿病肾病模型组、阳性药物组(吗替麦考酚酯胶囊,11 mg/kg)、金柚提取物组(25、50 mg/kg),8周后测定各组大鼠血尿素氮(BUN)、血肌酐(Scr)、24 h尿蛋白并留取肾组织。Western blot测定肾皮质中p-p38MAPK及胞核中NF-κB P65蛋白表达;酶联免疫吸附(ELISA)法测定血清白细胞介素8(IL-8)、转化生长因子-β(TGF-β)、纤维连接蛋白(FN)、纤溶酶原激活物抑制剂-1(PAI-1)等含量。结果:与正常对照组比较,糖尿病肾病模型组大鼠血清BUN、Scr含量,24 h尿蛋白,IL-8、TGF-β、FN、PAI-1水平、肾组织p-p38MAPK及胞核中NF-κB P65蛋白表达均显著升高(P<0.05)。经过治疗后,与糖尿病肾病模型组比较,阳性药物组和金柚提取物组(25、50 mg/kg)大鼠血清中各指标含量及各蛋白表达量均显著降低(P<0.05)。结论:金柚提取物对大鼠糖尿病肾病具有保护作用,其作用机制可能与调控肾脏组织中p38MAPK/NF-κBP65信号通路、减轻炎症反应有关。

    Effect of Extracts from Golden-pomelo on Expression of p38 Mitogen-activated Protein Kinase in Kidney of Diabetic Nephropathy Rats/ZHONG Zhihua,KANG Zhanfang.//Medical Innovation of China,2018,15(08):032-036

    【Abstract】 Objective:To study the effect of extracts from golden-pomelo on expression of p38 mitogen-activated protein kinase(p-p38MAPK) and nuclear factor κB P65(NF-κB P65) in the kidney of diabetic nephropathy rats.Method:The diabetic rats were induced by a single intraperitoneal injection of Streptozotocin.The rats were randomly divided into normal control group,diabetic nephropathy model group,positive drug group and extracts from golden-pomelo treatment group.The blood urea nitrogen(BUN),serum creatinine(Scr) and 24 h urine protein of each group were measured.The content of p-p38MAPK and NF-κB P65 was determined by Western blot.The serum interleukin 8(IL-8),transforming growth factor-β(TGF-β),fibronectin(FN) and fibrinolytic enzyme activators inhibitor 1(PAI-1) were detected with the ELISA method.Result:Compared with the normal control group,BUN,Scr,24 h urine protein,IL-8,TGF-β,FN,PAI-1,p-p38MAPK and NF-κB P65 protein of the kidney in the diabetic nephropathy model group were obviously enhanced(P<0.05).Compared with the diabetic nephropathy model group,BUN,Scr,24 h urine protein,IL-8,TGF-β,FN,PAI-1,p-p38MAPK and NF-κB P65 protein of the kidney in positive drug control group and extracts from golden-pomelo treatment group were significantly reduced(P<0.05).Conclusion:The extracts from golden-pomelo can protect diabetic rats kidney possibly by regulation p38MAPK/NF-κB signaling pathway to anti-inflammatory mechanism.
