PEDF抑制肝癌发生发展的作用机制研究(1)

     【摘要】 目的:探究色素衍生上皮因子(PEDF)在肝癌組织中表达的临床意义。方法:选取2013年1月-2017年10月本院接诊的80例肝癌患者为肝癌组及80例健康人作为对照组。检测比较两组及肝癌不同分期PEDF、血管内皮生长因子(VEGF)、人血小板反应蛋白1(TSP-1)、基质金属蛋白酶9(MMP-9)和微血管密度(MVD)、瘤坏死因子-α(TNF-α)、白细胞介素2(IL-2)、血栓调节蛋白(TM)、白细胞介素8(IL-8)水平,分析各指标间的相关关系。结果:肝癌组VEGF、MMP-9、TNF-α、TM、IL-8及MVD水平均高于对照组,而PEDF、TSP-1、IL-2水平均显著低于正常肝组织,比较差异均有统计学意义(P<0.05);VEGF、MMP-9、TNF-α、TM、IL-8和MVD水平随分化程度的下降而呈升高趋势,而PEDF、TSP-1和IL-2水平随分化程度的下降而呈下降趋势,比较差异均有统计学意义(P<0.05);PEDF与VEGF、TM和MVD呈负相关(P<0.05),与TSP-1呈正相关(P<0.05);VEGF与MMP-9、TM、MVD呈正相关(P<0.05),与TSP-1呈负相关(P<0.05);TSP-1与MVD、TM呈负相关(P<0.05);MMP-9、IL-8与MVD呈正相关(P<0.05),IL-2与MVD呈负相关(P<0.05);MMP-9与TM呈正相关(P<0.05)。结论:PEDF和TSP-1可能通过拮抗VEGF和MMP-9对肿瘤血管生成的调节作用来抑制TM和MVD,从而起到抗癌的作用,其在肝癌的治疗方面必将有广泛的用途。

    【Abstract】 Objective:To investigate the clinical significance of pigment derived epithelial factor(PEDF) expression in liver cancer tissue.Method:A total of 80 patients with liver cancer were selected as liver cancer group and 80 healthy people as control group from January 2013 to October 2017 in our hospital were selected.The levels of PEDF,vascular endothelial growth factor(VEGF),human platelet reactive protein 1(TSP-1),matrix metalloproteinase 9(MMP-9),microvessel density(MVD),tumor necrosis factor-α(TNF-α),interleukin 2(IL-2),thrombomodulin(TM),interleukin 8(IL-8) in different stages of liver cancer and two groups were compared,and analyze the correlation between the indicators.Result:The levels of VEGF,MMP-9,TNF-α,TM,IL-8 and MVD in liver cancer group were higher than those of control group,while the levels of PEDF,TSP-1 and IL-2 were significantly lower than those of control group,the differences were statistically significant(P<0.05).The levels of VEGF,MMP-9,TNF-α,TM,IL-8 and MVD increased with the decrease of differentiation,while the levels of PEDF,TSP-1 and IL-2 decreased with the decrease of differentiation,the differences were statistically significant(P<0.05).PEDF was negatively correlated with VEGF,TM and MVD(P<0.05),and positively correlated with TSP-1(P<0.05).VEGF was positively correlated with MMP-9,TM and MVD(P<0.05),and negatively correlated with TSP-1(P<0.05).TSP-1 was negatively correlated with MVD and TM(P<0.05).MMP-9 and IL-8 were positively correlated with MVD(P<0.05),and IL-2 was negatively correlated with MVD(P<0.05).MMP-9 was positively correlated with TM(P<0.05).Conclusion:PEDF and TSP-1 may inhibit TM and MVD by antagonizing the regulatory effect of VEGF and MMP-9 on tumor angiogenesis and it may play a role in anticancer.It will be widely used in treatment of liver cancer., 百拇医药(张鸿晖 刘清泉 熊秋生)
