白藜芦醇对宫颈癌Hela细胞VEGF表达和增殖的影响(1)
【摘要】目的 探讨白藜芦醇在体外对宫颈癌Hela细胞VEGF表达的影响。方法 MTT法检测不同浓度( 0、12.5、25、50、100、200、300、400、600 μmol/L) Res处理24、48、72h对HeLa细胞的抑制率;RT-PCR方法检测VEGFmRNA表达水平,Western Blot方法检测细胞内VEGF蛋白表达水平。结果 白藜芦醇呈时间剂量依赖性抑制宫颈癌Hela细胞的增殖;宫颈癌Hela细胞高水平表达VEGF,白藜芦醇能显著降低宫颈癌Hela细胞VEGF mRNA和蛋白表达。结论 白藜芦醇可以下调宫颈癌Hela细胞VEGF的表达,抑制宫颈癌细胞的增殖。【关键词】宫颈癌 血管内皮生长因子 白藜芦醇 增殖

【Abstract】Objective To investigate whether resveratrol could inhibit the vascular endothelial growth factor (VEGF) expression of human cervical cancer Hela cells in vitro. Methods Proliferation inhibitory effect was tested by MTT assay; VEGF mRNA was measured semi-quantitatively by RT-PCR, and the protein level was deteced by Western Blot.Results Resveratrol could inhibit the proliferation of Hela cells, with dose and time-depend effects; The level of VEGF was siginificantly decreased on Hela cells after incubated with resveratrol. Conclusion Resveratrol could inhibit proliferation and lower the expression of VEGF in human cervical cancer Hela cell line in vitro. ......
