核酸血筛标本冷链运输保存的方法和效果分析(1)

     [摘要] 目的 探讨血站的核酸标本冷链运输保存的方法和效果。方法 按照《血站技术操作规程(2015版)中标本运输的要求,模拟在车辆运输的条件下,使用不同的保存方法,测试运输箱内标本的保温时间(即标本运输温度保持在2~10℃的时间)。研究共分3组,第1组:标本运输箱底部放置一块4℃冷源板;第2组:标本运输箱底部放置一块冷源板,四周随机选取两面各放一块冷源板,共三块冷源板;第3组:标本运输箱底部放一块冷源板,四周各放一块冷源板,共五块冷源板。结果 模拟车内开空调环境,调节实验室内温度为23~27℃,湿度为30%~70%,第1、2、3组运输箱保温时间分别为:(0.16±0.02)h、(6.16±0.37)h和(13.78±0.71)h,两两比较差异均有统计学意义(F=2197.474,P<0.05)。结论 一块冷源不能满足核酸标本运输的要求,摆放3块冷源和5块冷源,可以确保核酸标本运输保存的温度在适宜范围内,为输血安全提供保障。

    The method and effect analysis of cold chain transportation of

    nucleic acid sieve

    [Abstract] Objective To investigate the ways and effects of blood sample cold chain transportation preservation in the blood center. Methods According to the requirements of Blood Stations Technology Operation Procedures (2015 edition), the holding time of the samples in the transport cases (the time of samples transport temperature holding between 2 to 10℃) were tested by different preservation methods, simulated the condition of transportation by vehicles. The samples were divided into three groups. The first group were used one piece of cold source plate at the bottom of the transport cases .The second groups were used three pieces of cold source plate, one piece were placed at the bottom, and the others were placed at two sides selected randomly. The third groups were used five pieces, one placed at the bottom, and the others were placed at each side. Results To simulate the condition in vehicles with air condition to regulate the temperature of a laboratory to 23-27℃, humidity to 30%-70%,the holding time in the first, the second and the third groups were (0.16±0.02)h、(6.16±0.37)h and (13.78±0.71)h, respectively. There was significant difference between the two groups (F=2197.474, P<0.05). Conclusion One piece of cold source cannot reach the requirement of nucleic acid samples transportation. Three and five pieces can ensure that the temperature of the transportation of nucleic acid samples is within the appropriate range, which can guarantee the safety of blood transfusion.

    目前我國血站输血相关传染性疾病的血液筛查普遍采用酶联免疫吸附实验(ELISA)方法,该方法的灵敏度和特异性虽然在不断地改进和提高,但由于方法学本身的局限性,感染者“窗口期”献血、病毒变异、隐匿性感染及其他因素仍然可能引起输血后HBV、HCV和HIV等病原微生物感染[1]。核酸扩增检测技术(nucleic acid amplification technologies,NAT)可以缩短病毒特异抗原和抗体免疫测定的“窗口期”[2-3],是提高血液安全的有效措施[4-7]。但是核酸检测与ELISA相比在检测过程的各个环节上均有其特殊之处,这些环节若缺乏严格的质量控制,会导致血液核酸检测结果不准确。, 百拇医药(么俊卿 叶惠芬)
